Analysis of autophagy in prostate tumor cells after treatment with selected cytostatics

Abstract:

Nádorová onemocnění prostaty v současné době patří mezi nejčastější malignity mužské populace. Tyto nádory se v pokročilých stádiích často vyznačují silnou rezistencí k terapii a špatnou prognózou. Autofagie, buněčný katabolický proces recyklace a odstraňování poškozených částí buněk, je nástrojem udržování buněčné homeostázy a brání nádorové transformaci. V již transformovaných nádorových buňkách ale může přispívat k jejich rezistenci, přežívání a napomáhat vzniku metastáz. V současnosti je tak předmětem výzkumu možnost farmakologické inhibice autofagie za účelem omezení jejího ochranného působení a posílení cytotoxického efektu terapie. V této práci jsme se u buněčných linií PC-3 a 22Rv1 testovali toxicitu běžně používaných inhibitorů autofagie 3-methyladeninu a Bafilomycinu A1, jejich vliv na autofagii a působení při kombinaci s cisplatinou. Zároveň jsme analyzovali expresi vybraných genů spojených s regulací autofagie a apoptózy. Naše výsledky ukázaly výrazné rozdíly v reakci na působení látkami mezi jednotlivými liniemi. Tato práce také ukazuje problémy spojené s látkami používanými v současné době pro experimentální ovlivňování autofagie, jejichž specifita není dostatečná a které silně ovlivňují celý metabolismus buněk.

Abstract:

Prostate cancer is currently one of the most common malignancies among male population. Prostate tumors in advanced stages are often characterized by strong resistance to therapy and by poor prognosis. Autophagy, a cellular catabolic process of recycling and removal of the damaged parts of the cells, is instrumental in maintaining of the cellular homeostasis and prevents tumor transformation. However, in already transformed cells, autophagy may promote their resistance, survival and metastatic activity. Recently, the possibility of pharmacological inhibition of autophagy to suppress its protective effect and to enhance the cytotoxic effects of the therapy has been studied extensively. This thesis focuses on the toxicity of commonly used autophagy inhibitors 3-methyladenine and Bafilomycin A1, their effect on autophagy and their effect on PC-3 and 22Rv1 cell lines when combined with cisplatin treatment. Our results show significant differences in response to the treatment between the cell lines. We also point out problems connected with the use of the tested autophagy inhibitors, which are nonspecific in their effects and affect the whole metabolism of the cells.
