Abstract:

Předkládaná disertační práce se zabývá některými aplikacemi hmotnostní spektrometrie v analytické chemii, jmenovitě: (i) stanovením antibiotik metodou LC-MS/MS po jejich předchozí extrakci z pevných organických vzorků a (ii) aplikacemi MALDI - a LDI-TOF-MS v analýze povrchu křemíkových waferů, identifikaci klastrů a analýze organických látek. Byl vyvinut nový selektivní a robustní postup stanovení vybraných antibiotik (tetracyklinů a sulfonamidů) v biologických vzorcích extrakcí kapalinou za zvýšeného tlaku (pressurized liquid extraction, PLE) a kapalinovou chromatografií spojenou s tandemovou hmotnostní spektrometrií (LC-MS/MS). Antibiotika byla extrahována metodou PLE a získané extrakty byly přečištěny extrakcí pevnou fází (solid phase extraction, SPE). Dále byla provedena identifikace a kvantifikace sledovaných látek metodou LC-MS/MS v módu multiple reaction monitoring (MRM) s vysokými hodnotami výtěžnosti (recovery). Metoda byla aplikována na analýzu reálných vzorků z odebraných z čističek odpadních vod na území Španělska. Další část disertační práce se zabývá využitím MALDI- a/nebo LDI-MS, zejména Quadrupole Ion Trap Time-Of-Flight Mass Spectrometry (QIT-TOF-MS), pro analýzu různých nanomateriálů nebo nanovrstev, analýzu piperazinu a jeho derivátů a také syntézu fosfidů arsenu metodou laserové ablace (laser ablation synthesis, LAS). Výsledky získané monitorováním syntézy klastrů laserovou ablací nano-prekurzorů přispívají k pochopení vzniku chemických vazeb v laserem generovaných klastrech. Stechiometrie syntetizovaných As-P klastrů byla určena metodou izotopových obalových křivek (isotopic envelope analysis) a pomocí počítačových modelů. Ukazuje se, že hmotnostní spektrometrie je vhodná detekční metoda a lze ji ve spojení s vhodnou instrumentací (HPLC, MALDI/LDI) aplikovat nejen na analýzu organických látek, ale také na materiály anorganické, včetně nanomateriálů, a ke sledování produktů syntéz využívajících laserovou ablaci.

Abstract:

PhD Thesis deals with applications of mass spectrometry in analytical chemistry, namely: (i) LC-MS/MS determination of antibiotics after theirs extraction from biosolids and (ii) MALDI or LDI TOF MS applications for surface analysis of silicon wafers, clusters and organic compounds. New procedure for the determination of antibiotics (tetracycline’s and sulphonamide’s) in biosolids by pressurized liquid extraction (PLE) and liquid chromatography-tandem mass spectrometry was worked out. Robust and sensitive method for quantitative determination of selected antibiotics was developed. Antibiotics were extracted using PLE and clean-up of the extracts was carried out by solid phase extraction (SPE). Identification and quantification of the compounds by LC-MS/MS in multiple reaction monitoring (MRM) mode was done. High recoveries were obtained. The method was applied to the analysis of real sludge samples collected from different WWTPs in Spain. The Thesis also deals with MALDI and/or LDI MS, including Quadrupole Ion Trap Time-Of-Flight Mass Spectrometry (QIT-TOF-MS) analysis of various nano-materials or nano-layers and with laser ablation synthesis of arsenic phosphides, analysis of piperazine and its derivatives. Original results concerning the laser ablation synthesis of new As-P clusters s from nano – precursors were obtained and the results contribute to the understanding of chemical bonding in laser generated clusters. The stoichiometry of the laser ablation synthesized As-P clusters was determined via isotopic envelope analysis and computer modelling. Concluding, it is demonstrated that MS detection is advantageous for detection in coupling with HPLC, MALDI TOF MS instrumentation can be used with advantage not only for analysis of organic compounds but also for inorganic materials including nano and for laser ablation synthesis.
