The changes of intracellular lipids during induced differentiation and apoptosis of colon cells

Abstract:

Buňky tlustého střeva jsou vystaveny jak endogenním regulátorům proliferace, diferenciace a apoptózy, tak lipidovým složkám potravy, jako jsou vysoce nenasycené mastné kyseliny (VNMK) a mastné kyseliny s krátkým řetězcem (MKKR) – butyrát, který vzniká anaerobní mikrobiální fermentací vlákniny. Signály indukované těmito faktory jsou integrovány v buňce a mohou významně ovlivnit buněčný metabolizmus a kinetiku. Dynamická rovnováha mezi proliferací, diferenciací a apoptózu je pro zdraví střevního epitelu nezbytná a lipidové složky potravy mohou v zajištění homeostázy sehrát významnou roli. Procesy diferenciace a apoptózy lidských buněk jsou spojeny se změnami biofyzikálních vlastností buněčné membrány a buněčných lipidů. Změny ve složení, struktuře a metabolizmu buněčných lipidů se mohou významně lišit u buněk normálních a nádorových. V naší práci jsme srovnávali odpověď normálních nenádorových buněk fetálního střeva FHC a buněk karcinomu střeva HCT-116 na působení VNMK třídy n-3 (DHA, 50 microM) a n-6 (AA, 50 microM) a NaBt (3 mM). Hlavní použitou metodou byla průtoková cytometrie. Modulace proliferace (počty buněk, buněčný cyklus), buněčné smrt (% plovoucích buněk, sub G0/G1 populace, mitochondriálním membránový potenciál, morfologie jádra, štěpení PARP a exprese antiapoptotického proteinu Mcl-1) a diferenciace (aktivita ALP) byly srovnávány se změnami buněčných lipidů (akumulace TAG v cytoplazmě, změny ve struktuře lipidové membrány a produkce ROS).

Abstract:

Together with endogenous regulators of proliferation, differentiation and apoptosis, colon epithelial cells are exposed to dietary lipid compounds like esential polyunsaturated fatty acids (PUFAs) and short chain fatty acid - butyrate (produced by microbial fermentation of fibre). Signals induced by these factors are integrated inside the cells and may have significant effects on cell metabolism and kinetics. Dynamic balance between proliferation, differentiation and apoptosis is necessary for healthy intestinal epithelium and lipid compounds can play an important role in maintaining homeostasis in this tissue. Processes of differentiation and apoptosis of human cells are accompanied with changes of biophysical properties of cell membranes and intracellular lipids. Modulation of constitution, structure, symmetry and metabolism of cell lipids may be different in normal and tumor cells. Detection of these alterations and their correlation with other parameters reflecting differentiation and apoptosis contribute to clarifying of sequencing and regulation of particular steps of these processes and differences between normal and tumor cells. We compared the response of human colonic cell lines derived from normal fetal tissue (FHC) and colon adenocarcinoma (HCT116) to treatment with sodium butyrate (NaBt, 3 mM) and PUFAs (50 microM) of n-6 (arachidonic acid, AA) or n-3 (docosahexaenoic acid, DHA) series. Flow cytometry was the main methodology used. While in normal FHC cells NaBt induced differentiation, in HCT116 cancer cells it induced apoptosis. Combination of NaBt with AA or DHA attenuated FHC cell differentiation and strongly enhanced apoptotic response. No such effects were observed in HCT116 cells. Modulations of proliferation (cell number, cell cycle), differentiation (activity of alkaline phosphatase), and cell death (% of floating cells, subG0/G1 population, mitochondrial membrane potential - TMRE fluorescence, nuclear morphology, cleavage of PARP and expression of antiapoptotic protein Mcl-1) were associated with changes of membrane lipid packing (merocyanine 540 fluorescence), accumulation of lipid droplets in cytoplasm (Nile red fluorescence), and reactive oxygen species production (dihydrorhodamine-123 fluorescence). Our results suggest that NaBt as well as PUFAs may cause significant changes in colonic cell lipids, influence oxidative metabolism and modulate colonic cell kinetics differently in normal and tumor cells.
