Function of the antimicrobial peptides in humoral immune system of honeybees (Apis mellifera)

Abstract:

Antimikrobiální peptidy představují důležitou složku nespecifické imunity včely medonosné. Jsou syntetizovány v první vlně imunitní odpovědi organismu a jsou schopny rychle a účinně interagovat s patogenními mikroorganismy, jako jsou bakterie, kvasinky, či viry. Jedním z hlavních témat této práce je studium indukce tvorby antimikrobiálních peptidů v organismu včely při infekci a jejich ovlivnění z hlediska výživy. V experimentálních studiích první části práce byly sledovány parametry imunitního systému (kvantifikace peptidu apidaecinu, stanovení relativní genové exprese antimikrobiálních peptidů apidaecinu, abaecinu, defensinu-1, defensinu-2, hymenoptaecinu, koncentrace a relativní genová exprese zásobního proteinu vitellogeninu a nutričních proteinů mrjp) v odpovědi na injekci směsi G+ a G- bakterií u dvou generací včel a při dlouhodobé aplikaci spor patogenů do krmiva mladých včel starajících se o včelí larvy. Bylo zjištěno, že u zimní generace včel dochází v odpovědi na injekci bakterií k mnohem intenzivnější aktivaci imunitního systému než u letní generace. Aktivace imunity u zimních včel také vede k poklesu hladiny a relativní genové exprese zásobního proteinu vitellogeninu. Výrazná aktivace imunity u zimní generace včel tak probíhá na úkor spotřeby důležitých zásobních proteinů. V druhé experimentální studii byla po orální aplikaci spor patogenů do krmiva včel po dobu 10 dní detekována zvýšená exprese antimikrobiálního peptidu apidaecinu a zásobních proteinů mrjp 1 a mrjp 3. Toto zvýšení je podmíněno přídavkem pylu do krmiva včel. Dostatek proteinů ve výživě včel má vliv jak na tvorbu humorálních faktorů, tak i na tvorbu nutričních proteinů MRJP. Druhý experimentální celek práce se zbývá využitím řasy Chlorella vulgaris jako potenciálního doplňku potravy včel. Byla studována antimikrobiální aktivita extraktů řasy Ch. vulgaris vůči včelí patogenní bakterii Paenibacillus larvae. U několika frakcí nejvíce aktivního extraktu byla stanovena minimální inhibiční koncentrace vůči vegetativním bakteriím i sporám P. larvae a identifikovány sloučeniny zodpovědné za antimikrobiální aktivitu (kys. linolenová, monolinolenin a monolinolein). Využití řasy jako proteinové náhražky bylo dále zhodnoceno studiem vlivu konzumace řasy Ch. vulgaris na antioxidační systém včel (aktivita antioxidačních enzymu, relativní genová exprese genů antioxidačních enzymů) a hladinu zásobního proteinu vitellogeninu v hemolymfě včel. Po konzumaci řasy vzrůstá hladina proteinů a zásobního proteinu vitellogeninu v hemolymfě včel, ale není stimulována genová exprese vitellogeninu. Nebyla pozorována vyšší aktivita trávicích enzymů ani antioxidačních enzymů v hemolymfě včel. Výsledky experimentů této disertační práce zaměřených na imunitní systém včely poskytují pevný základ pro další výzkum, který bude zaměřen na přesný časový průběh produkce humorálních faktorů od imunostimulu a na studium aktivace imunity na úrovni signálních drah. Druhý celek práce přináší nový přístup ke zhodnocení potenciálních potravinových doplňků využitelných ve včelaření a přináší prvotní zhodnocení řasy Ch. vulgaris při jejím využití jako krmného sirupu.

Abstract:

Antimicrobial peptides play an important role in the honey bee's non-specific immunity. They are synthesized in the first line of the immune response in the organism and can quickly and effectively interact with pathogenic microorganisms such as bacteria, yeasts, or viruses. One of the main aims of this work was to study the induction of the production of these peptides in the honey bee organism during infection and the influence in their production in terms of nutrition. In experimental studies, immune system parameters were studied (quantification of the peptide apidaecin, determination of the relative gene expression of antimicrobial peptides apidaecin, abaecin, defensin-1, defensin-2, hymenoptaecin, concentration, and relative gene expression of the storage protein vitellogenin and the nutritional proteins mrjp) in response to the injection of a mixture of G+ and G- bacteria in two generations of bees and during long-term application of pathogen spores to the food of nurse bees. It was observed that the winter population of bees has a more intense activation of the immune system in response to the injection of bacteria than the summer population. The intense activation of immunity in winter bees resulted in a decrease in the level and relative gene expression of the important storage protein vitellogenin. In the second experimental study, increased expression of the antimicrobial peptide apidaecin and the storage proteins mrjp 1 and mrjp 3 was detected after oral application of pathogen spores to nurse bees for 10 days. This increase was dependent on the addition of pollen to the bee feed. Therefore, proteins in bee nutrition play an important role in the production of humoral factors and nutrition proteins MRJP. The second part of the experimental work was focused on the use of the alga Chlorella vulgaris as a potential food supplement for honey bees. The antimicrobial activity of Ch. vulgaris algae extracts against the bee pathogenic bacterium Paenibacillus larvae was studied. Minimum inhibitory concentrations against vegetative bacteria and P. larvae spores were determined for several fractions of selected extract and the compounds responsible for the antimicrobial activity (linolenic acid, monolinolenic acid, and monolinoleic acid) were identified. The use of alga as a protein supplement was further evaluated by studying the effect of Ch.vulgaris consumption on the antioxidant system of bees (antioxidant enzyme activity, relative gene expression of antioxidant enzymes genes) and at the level of the storage protein vitellogenin in the haemolymph of bees. After consumption of algae, the protein concentration, and the level of storage protein vitellogenin increased in the hemolymph of bees. The relative gene expression of vitellogenin was not stimulated. No higher activity of digestive enzymes or antioxidant enzymes was observed in the hemolymph of bees. The experimental work in this study focused on the bee immune system provides a solid base and guidelines for further research that will focus on the time-dependent production of humoral factors from immunostimulation and on the precise activation of immunity at the level of signaling pathways. The second part of this thesis provides a novel approach to the evaluation of potential food supplements useful in beekeeping and provides an initial evaluation of the alga Ch. vulgaris when used as a feed syrup.
