New approaches for genotoxicity assessment in ecotoxicology

Abstract:

V této bakalářské práci se věnuji aktuálním tématům, týkajícím se metod testování genotoxického působení látek. Zabývám se okrajově současnými trendy výzkumu a uvádím přehled nejpoužívanějších testů – In vitro mikronukleus test na savčích buňkách, Amesův test, Test chromozomálních aberací, In vitro test genových mutací na savčích buňkách, Umu test. Samotná práce je zaměřena na nové metody a vylepšení metod stávajících – Model lidské kůže EpiDermTM, In vitro test deleční rekombinace DNA u kvasinek, Mikronukleus test a jeho stanovení průtokovou cytometrií, Cda genotox test, GADD45α-GFP GreenScreen HC. V praktické části práce se zabývám ověřením použitelnosti testovacího modelu GreenScreen GC prováděného na kvasinkových kulturách.

Abstract:

In this thesis I follow current issues in testing methods of genotoxic effects of substances. Marginally I deal with actual trends in research and give an overview of the most widely used assays – In vitro mammalian cell micronucleus test, Ames test, Chromosome aberration test, In vitro mammalian cell gene mutation test, Umu test. The thesis itself is focused on new methods and improvements to existing methods - Human skin model EpiDermTM, In vitro yeast DNA del recombination assay, In vitro flow cytometry micronucleus assay, Cda genotox assay, GADD45α-GFP GreenScreen HC. In the practical section of this paper I deal with verifying the applicability of the GreenScreen GC assay, which is carried out on yeast cultures.
