Genetic and morphological variability of tapeworm Khawia armeniaca (Cholodkovski, 1915) (Cestoda: Caryophyllidea)

Abstract:

Tasemnice Khawia armenica (Caryophyllidea) parazitující ve sladkovodních rybách z čeledi Cyprinidae, byla od svého popisu v roce 1915 zaznamenána v řadě zemí Afriky, Asie a Evropy. Tento druh byl v porovnání s ostatními zástupci rodu tradičně považován za zástupce s nejširším hostitelským spektrem a největším zeměpisným rozšířením, které zahrnuje dvě zoogeografické oblasti (tj. Palearktickou a Etiopskou). Nedávná taxonomická revize rodu Khawia však v rámci druhu K. armeniaca odhalila významnou vnitrodruhovou variabilitu, což vedlo k předpokladu, že se pravděpodobně jedná o komplex druhů. Předložená diplomová práce si klade za cíl prozkoumat tuto diverzitu pomocí morfologické a molekulární analýzy vzorků získaných z 13 druhů cyprinoidních a 1 druhu mastacembelidních ryb z oblasti Blízké-ho východu, severní Afriky a Pyrenejského poloostrova. Pro posouzení molekulární diverzity druhu K. armeniaca byly sekvenovány úseky 18S rDNA, 28S rDNA a oblast ITS2. Provedená fylogenetická analýza rozdělila všechny zkoumané jedince do dvou linií, které jednoznačně odpovídaly jejich geografickému původu. Zástupci první linie z hostitelů Arabibarbus grypus, Barbus lacerta, Capoeta birunii, Carassobarbus lu-teus, Luciobarbus barbulus, L. esocinus a L. kersin z Iráku a Íránu jsou geneticky identické s jedinci Khawia armeniaca z typové lokality druhu, jezera Sevan v Arménii. Tasemnice druhé linie byly nalezeny u Luciobarbus bocagei, L. comizo a L. guiraonis z Portugalska a Španělska. Morfologická analýza založená na 40 morfometrických znacích ukázala, že se obě linie liší tvarem, strukturou a velikostí vaječníku a rozsahem žloutkových folikulů v přední části těla. Na základě těchto molekulárních i morfologických rozdílů je linie tasemnic z Pyrenejského poloostrova považována za nový druh.

Abstract:

Caryophyllidean tapeworm Khawia armeniaca, parasite of freshwater fishes of the family Cyprinidae, has been reported from various countries in Africa, Asia and Europe since its initial description in 1915. This tapeworm was originally assumed to represent a species with the widest host spectrum among all the other congeneric species, along with the broadest geographical distribution spanning two zoogeographical regions (i.e., the Palearctic and Ethiopian). However, a recent taxonomical revision of the genus Khawia revealed significant intraspecific variability within K. armeniaca, which led to an assumption that it might actually constitute a complex of species. This thesis aims to investigate this diversity by analyzing samples extracted from 13 cyprinid and 1 mastacembelid species in the Middle East, Northern Africa and the Iberian Peninsula, using both molecular and morphological approach. To assess the molecular diversity of K. armeniaca, partial 18S rDNA, 28S rDNA, and ITS2 region were sequenced. The conducted phylogenetic analysis divided all investigated individuals into two distinct lineages, which clearly corresponded to their geographical origin. The tapeworms of the first lineage from Arabibarbus grypus, Capoeta birunii, Barbus lacerta, Carassobarbus luteus, Luciobarbus barbulus, L. esocinus, and L. kersin in Iraq and Iran are genetically identical to Khawia armeniaca, which was described from the Sevan khramulya Capoeta sevangi from the type locality of lake Sevan in Armenia. Tapeworms of the other lineage are found in Luciobarbus bocagei, L. comizo, and L. guiraonis from Portugal and Spain. Morphological analyses on a total of 40 morphometric characters revealed that the tapeworms of both lineages mainly differ from each other in shape, structure and size of the ovary and the anterior extent of the vitelline follicles. Upon comprehensive evaluation of both molecular and morphological distinctions, the tapeworms from the Iberian Peninsula are regarded as a new species.
