The application of microcalorimetry in the study of enzymatic reactions

Abstract:

Cílem této práce je studium kinetiky enzymatické reakce trypsinu a substrátu N?-bezoyl-L-arginin-p-nitroanilid hydrochloridu (BApNA) metodou mikrokalorimetrie a spektrofotometrie a nalezení takových podmínek experimentu, aby akceptovaly omezení použitých technik a vyhovovaly požadavkům na měření. Současně byla sledována kinetika reakce trypsinu a substrátu BApNA v závislosti na změně reakčních podmínek, jako byla změna teploty, koncentrace enzymu, koncentrace substrátu a poměru látkových množství enzymu a substrátu.

Abstract:

The aim of this diploma work is the study of the kinetics of enzymatic reaction of trypsin and its substrate N?-bezoyl-L-arginine-p-nitroanilid hydrochloride (BApNA) using microcalorimeter and spectrophotometer and to discover such conditions of the experiment which would accept a limitation of used techniques and meet the requirements for the measurement. The kinetics of reaction of trypsin and its substrate BApNA was studied as well as its dependence on the reaction conditions like changes in temperature, enzyme concentration, substrate concentration and the ratio of the quantity of enzyme and substrate was also monitored.
