Detection of genes responsible for the blue color in common wheat (Triticum aes-tivum) grain

Abstract:

Pšenice setá je jedna z nejvýznamnějších zemědělských plodin, je hojně pěstována po celém světě a samozřejmě i v České republice. V posledních letech stále více roste zájem o pěstování pšenic s nestandardně zbarveným zrnem – jedná se nejčastěji o purpurové, modré a žluté pšenice. Tyto vyšlechtěné kultivary obsahují ve větší míře zdraví prospěšné látky, jako jsou antokyany a karotenoidy. V případě modrých pšenic projev modrého zbarvení kódují Ba geny (blue aleuron), konkrétně Ba1, Ba2 a Ba3 geny. Pro první dva jmenované byly objeveny kandidátní geny, ThMYC4E pro Ba1 gen a TbMYC4A pro Ba2 gen, které zajišťují jejich expresi, a tím zbarvují aleuron do modra. Tato bakalářská práce se zaměřila na možnost testování jednotlivých kultivarů na přítomnost těchto markerů pomocí jednoduché metodiky založené na PCR reakci a sekvenování PCR produktů. Analýzy byly doplněny o extrakci antokyanů z variant s modrým zrnem a také o fylogenetické vyhodnocení. Prezentované výsledky dokazují přítomnost těchto markerů ve vybraných kultivarech a využitá metodika tak může sloužit pro testování barevných variant pšenice seté v praxi.

Abstract:

Common wheat (Triticum aestivum) is one of the most important agricultural crops, it is widely grown all over the world and of course in the Czech Republic. In recent years, there has been a growing interest in the production of non-standard grain coloration wheat cultivars, most often purple, blue and yellow wheat cultivars. These cultivars contain a higher concentration of health-promoting substances such as anthocyanins and carotenoids. In the blue aleurone wheat cultivars, the expression of blue colour is encoded by the Ba (blue aleurone) genes – namely Ba1, Ba2 and Ba3 genes. For the Ba1 and Ba2 genes, there have been discovered candidate genes, ThMYC4E for Ba1 gene and TbMYC4A for Ba2 gene, which control the expression of the Ba genes and as consequence aleurone changes its colour to blue. This bachelor thesis was focused on the testing blue wheat cultivars for the presence of these markers using a PCR reaction and sequencing of the PCR products. The analyses were complemented by the anthocyanins extraction from the blue aleurone cultivars and by phylogenetic analysis. Results demonstrate the presence of these markers in the selected cultivars and used methodology can be used for the testing of colored common wheat cultivars in practise.
