Duplex sequencing detects ultra-rare mutations by tagging DNA molecules with double-stranded tags. This method creates single-stranded consensus sequences (SSCS) from the reads, which then form duplex consensus sequences (DCS) and are then aligned to the reference genome to call mutations.
