Mutation detection in hereditary cancer

Abstract:

Tato práce se zabývá vznikem mutací v genech, jejich detekcí a následným vznikem rakovinového procesu. Současně se věnuje následkům těchto patogenních změn v lidském organismu. Větší důraz je kladen na detekční techniky. Jsou zde zahrnuty jak metody, které stály na počátku vzniku této oblasti diagnostiky, dále nejpoužívanější a nejcitlivější metody, tak i metody moderní. Kromě této teoretické části vysvětlující vznik mutací jako patologické změny v organismu, procesu rakovinového bujení a popisu analytických detekčních postupů, tato práce obsahuje také příklad nádorového onemocnění, na kterém je demonstrována celá teorie a dále strategický postup selekce jednotlivých případů pacientů dle jejich závažnosti. Současně tato práce obsahuje přílohu, ve které jsou uvedeny statistické údaje vybraného nádorového onemocnění od roku 1977 - 2007. Tyto údaje ukazují stav incidence tohoto onemocnění v ČR a Pardubickém kraji z různých hledisek.

Abstract:

This thesis deals with mutation in genes, their detection and subsequent development of cancer process. At the same time, it shows the results of these pathogenic changes in a human body. More stress is put on detection techniques. The methods which stood in the beginning of the origin of this branch of diagnostics can be found here, further you can find the most frequent and most sensitive methods, and modern methods as well. Apart from this theoretical part explaining the origin of mutation as pathological changes in a body and dealing with the process of cancer proliferation and description of analytic detection procedures, this thesis includes an example of a tumour disease which the whole theory is demostrated on, and a strategic process of the selection of individual cases according to their seriousness. This thesis contains a supplement in which statistical data of a selected tumour disease from the year 1977 to 2007 are mentioned. These data show the state of incidence in the Czech Republic and Pardubice region from various points of view.
