Screening of late defence reaction on tobacco in dependence on applied elicitor

Abstract:

Kryptogein je proteinový elicitor dekretovaný oomycetou Phytopthtora cryptogea. Patří mezi elicitiny a u rostlin tabáku vyvolává hypersenzitivní reakci. Kryptogein se váže na vysokoafinitní místa na plasmatické membráně a tím spouští depolarizaci plasmatické membrány, fosforylaci proteinů, alkalizaci extracalulárního média, výtok sodných a chloridových iontů z buňky a vtok vápenatých iontů do buňky a také přechodnou produkci aktivních forem kyslíku. Až několik hodin po elicitaci nastává syntéza PR proteinů a indukce lokální rezistence.Tato práce je zaměřena na studium vztahu mezi strukturou elicitinu a jeho schopností vyvolat obrannou reakci u rostlin. Nejprve jsem připravila mutantní formu kryptogeinu K13V, který má v pozici 13 zaměněn lysin za valin. U tohoto proteinu byla, s použitím dehydroergosterolu (DHE), testována jeho schopnost vázat steroly. Prokázalo se, že protein K13V je schopen vázat steroly, ale oproti divokému typu má ke sterolům nižší afinitu.Dále jsem sledovala schopnost mutantních forem kryptogeinu vyvolat obrannou reakci u rostlin. Pro tyto účely byl použit divoký typ kryptogeinu, protein K13V a již dříve připravené mutantní formy kryptogeinu L41F, V84F, V84FL41F. Listy rostlin tabáku pak byly inokulovány 5nM roztokem elicitoru. A byla zkoumána jejich schopnost vyvolat nekrózu, syntézu obranných proteinu a syntézu fytoalexinu kapsidiolu. Nejsilnější nekróza se projevila u listů rostlin ošetřených proteinem X24 a V84F, slabší pak byla u proteinu K13V. V případě proteinů L41F a V84FL41F nebyla nekróza žádná. Indukce nekrózy korelovala s expresí genů pro enzymy tPOX a APX, které detoxifikují H2O2 a STC, která hraje klíčovou roli při syntéze fytoalexinu kapsidiolu. Byla nalezena korelace mezi expresí genu PR1a proteinu a expresí genu pro enzym PAL, který je důležitým regulačním enzymem fenylpropanoidové dráhy vedoucí k produkci signální molekuly kyseliny salicylové, která se podílí na regulaci exprese kyselých forem PR proteinů. V rámci tohoto experimentu byla dále sledována exprese genů pro proteiny PR-1b, PR-3Q a PR-5. Jednotlivé PR proteiny vykazovaly různý charakter exprese, což je zřejmě způsobeno zapojením rozdílných regulačních mechanismů podílejících se na indukci exprese PR proteinů.

Abstract:

Cryptogein is a proteinaceous elicitor secreted by oomyceta Phytopthtora cryptogea. It belongs to elicitin family and induces a hypersensitive response on tobacco cells. Cryptogein interacts with high affinity binding site on plasma membrane and triggers depolarization of the plasma membrane, protein phosphorylation, and alkalization of extracellular medium, calcium and chloride ions efflux and large calcium influx and transient synthesis of AOS in tobacco. Several hours after elicitation the expression of PR proteins and induction of local resistance is observed. This thesis is focused on relationship between structure of elicitins and their ability to induce plants defence reaction. Firstly, I prepared a new kryptogein mutant K13V, which carries substitution of lysin on position 13 for valin. Using DHE, a fluorescent sterol, I demonstrated that K13V interact with sterol but with lower affinity than wild type X24. Consequently, I tested the capability of cryptogein mutants to induce plants defence reaction. For this study wild-type cryptogein X24, K13V mutant and three previously prepared recombinant cryptogein mutants (L41F, V84F and V84F/L41F) were used. The leaves of tobacco plants were treated with 5nM solution of each cryptogein mutants and their ability to induce necrosis, synthesis of defence genes and synthesis of capsidiol was investigated. The strongest necrosis was observed in proteins X24 and V84F, less in K13V and none in L41F and V84F/L41F. This observation correlates with expression of genes for enzymes tPOX and APX, which detoxify H2O2 and STC, which is a key enzyme in synthesis of phytoalexin capsidiol. As well, correlation was found between the expression of gene for protein PR-1a and expression of gene for enzymes phenyalaninammonium lyase (PAL), which is a key regulation enzyme of phenylpropanoid pathway resulting in SA production playing an important role in regulation of acidic PR forms expression. Further, in this thesis expression of other PR proteins were studied, such as PR-1b, PR-3Q and PR-5. Different PR proteins showed different patterns of expression, which may indicate involvement of different mechanisms to induction of PR proteins expression.
