Immunocomptence and parasitism in fish species with the variable level of ploidy living in aquaculture and natural habitat

Abstract:

Předložená bakalářská práce se zabývá problematikou změn úrovně ploidie u ryb a jejím vztahem k parasitární infekci. První část práce řeší problematiku výskytu polyploidie u ryb, metody její detekce a její umělou indukci v akvakulturách. Práce je zaměřena především na rozdíly mezi diploidními a triploidními rybami. Poskytuje základní srovnání imunologických a fyziologických parametrů u 2n a 3n ryb a poukazuje na rozdíly mezi nimi. Modelovým hostitelem této práce je sladkovodní ryba lín obecný (Tinca tinca), který je obecně používán k indukci triploidie v akvakulturách. Druhá část zahrnuje přehled metod odlovu, pitvy, sběru, fixace, barvení a determinaci parazitů. Doposud byly provedeny tři odlovy ve Vodňanech a odloveno celkem 60 diploidních a 60 triploidních ryb v poměru pohlaví 1:1. Poslední část práce zahrnuje předběžné výsledky prvního odlovu 18 diploidních a 22 triploidních línů v září 2008. Bylo zde nalezeno 5 druhů parazitů: A. tincae (346 jedinců), V. campylancristrota (16), T. clavata (4), Caryophyllaeus sp. (2), G. tincae (2). Byly vyhodnoceny základní epidemiologické charakteristiky: prevalence, abundance a intenzita infekce. Nejvyšší prevalence pro 2n byla u A. tincae a pro 3n byla u V. campylancristrota. Intenzita infekce byla významně vyšší opět u A. tincae vyskytující se ve střevech u 2n línů. Větší diverzita parazitů je u 3n línů, ale u 2n je zase vyšší početnost.

Abstract:

This bachelor thesis deals with changes ploidy levels in fish and its relationship to parasite infection. The first part of the work deals with the occurrence of polyploidy in fishes, methods of detection and artificial induction in aquacultures. The work focuses on the differences between diploid and triploid fish. It provides a basic comparision of immunological and physiological parameters in 2n and 3n fish and highlights the differences between them. Model hosts of this work is common freshwater fish tench (Tinca tinca), which is commonly used for induction triploidy in aquacultures. The second part includes an overview of methods of harvesting dissection, collection, fixation, staining and determination of parasites. Three catches have been done in Vodňany and 60 diploid and 60 triploid fish in sex ratio 1:1. The last part of the work involves the preliminary results of the first catch of 18 diploid and 22 triploid tench in September 2008. There were found 5 species of parasites: A. tincae (346 individuals), V. campylancristrota (16), T. clavata (4), Caryophyllaeus sp. (2), G. tincae (2). Basic epidemiological characteristics were assessed: prevalence, abundance and intensity of infection. The highest prevalence for 2n was in A. tincae and for 3n was V. campylancristrota. The intensity of infection was significantly higher again in A. tincae occurring in the intestines of 2n tench. Greater diversity of parasites apears by 3n tench but by 2n is higher abundance.
