3D cultivation of mammary epithelial cells for the study of mammary gland pathogenesis

Abstract:

Tato diplomová práce se zabývá možnostmi kultivace epitelových buněk mléčné žlázy s využitím 2D a 3D kultivačních přístupů se zaměřením na tvorbu trojrozměrných struktur. Hlavním cílem bylo porovnat různé způsoby kultivace buněčné linie PS a zhodnotit jejich vhodnost pro tvorbu sféroidů s ohledem na studium fyziologie a patogeneze mléčné žlázy. V experimentální části byly buňky kultivovány ve 2D podmínkách a ve vybraných 3D modelech, konkrétně scaffold-based přístupy s využitím Matrigelu a scaffold-free metody hanging drop. Byla sledována morfologie buněk, jejich schopnost agregace a tvorba trojrozměrných struktur v závislosti na použitých kultivačních podmínkách. Výsledky naznačují, že scaffold-based modely s využitím Matrigelu podporují vznik kompaktních sféroidních struktur. Naproti tomu při využití metody hanging drop docházelo především k tvorbě buněčných agregátů bez typické sféroidní organizace. Získané poznatky naznačují, že volba kultivačního přístupu zásadně ovlivňuje vlastnosti vznikajících struktur a jejich potencionální využití. Vytvořené 3D modely mohou představovat vhodný přístup pro studium procesů probíhajících v mléčné žláze, včetně zánětlivých onemocnění, jako je mastitida.

Abstract:

This diploma thesis focuses on the cultivation of mammary epithelial cells using both 2D and 3D culture approaches, with an emphasis on the formation of three-dimensional structures. The main aim was to compare different culture methods of the PS cell line and to evaluate their suitability for spheroid formation with respect to the study of mammary gland physiology and pathogenesis. In the experimental part, the cells were cultured under 2D conditions and in 3D models, specifically scaffold-based approaches using Matrigel and a scaffold-free method based on the hanging drop technique. Cell morphology, aggregation ability, and the formation of three- dimensional structures were evaluated depending on the culture conditions used. The results suggest that scaffold-based models using Matrigel support the formation of compact spheroidal structures. In contrast, the hanging drop method primarily led to the formation of cell aggregates without typical spheroidal organization. The findings indicate that the choice of culture approach significantly influences the properties of the resulting structures and their potential applications. The developed 3D models may represent a suitable approach for studying processes occurring in the mammary gland, including inflammatory conditions such as mastitis.
