Transcriptomic analysis of astrocytes derived from iPSC cells of patients with sporadic form of Alzheimer's disease

Abstract:

Alzheimerova choroba je nejznámější neurodegenerativní onemocnění způsobující demenci. Navzdory tomu, že v současné době trpí tímto onemocněním zhruba 50 milionu lidí a očekává se, že v budoucnu tento počet poroste až trojnásobně, stále neexistuje žádná efektivní léčba, která by zvrátila toto onemocnění. Z těchto důvodů je snaha studovat příčiny a mechanismy, které by pomohly najít nové terapeutické cíle s nadějí úspěšné léčby. Značkou překážkou je to, že pro studium Alzheimerovy choroby chybí adekvátní modely, jež by poskytly potřebné informace. Nová metoda iPSC buněk se prozatím jeví jako nejadekvátnější řešení, neboť umožňuje kultivaci mozkových buněk in vitro při zachování patologického profilu jedince. V této diplomové práci byly použity iPSC buňky, ze kterých byly třemi způsoby diferencovány lidské astrocyty. Na základě transkriptomické analýzy bylo zjištěno, že in vitro kultivace astrocytů má sice do budoucna jistý potenciál, avšak k opravdovým dospělým astrocytům to má ještě hodně daleko a je vyžadováno další zkoumání.

Abstract:

Alzheimer's disease is probably the most well-known neurogenerative disorder leading to dementia. In spite of the fact there is currently approximately 50 million of people who have been suffering from this disease and it is expected that the number is about to increase three times in the following decades, a proper treatment enabling to reverse the process of illness still has not been found. With that being said, there is a strong urge to study the causes and mechanisms, which could contribute to find new therapeutic targets with the hope of successful treatment. The biggest obstacle here is that for observing Alzheimer's disease a proper models are lacking. Therefore, new method of an iPSC cells seems to be an appropriate solution since it allows in vitro cultivation of brain cells while preserving pathological profile of an individual. In this diploma thesis the iPSC cells were used for differentiation in three different ways to obtain human astrocytes. Based on the transcriptomic analysis it was discovered that in vitro cultured astrocytes have a certain potential for the future, yet it is still far away from real matured astrocytes, hence further exploration is required.
