The role of Sertoli cells in endocrine disruption

Abstract:

V posledních letech se do popředí zájmu odborné a široké veřejnosti dostávají údaje týkající se samčího reprodukční zdraví a narušení jeho funkcí. Existují epidemiologické studie, které spojují účinky konkrétních látek na reprodukční systém lidí i divoce žijících zvířat. Cílem této diplomové práce bylo sepsat literární rešerši týkající se role Sertoliho buněk v endokrinní disrupci a prostudovat účinky významných environmentálních polutantů přímo na Sertoliho buňky, klíčové somatické buňky varlat. Sertoliho buňky jsou nezbytné pro vznik a vývoj varlat a tvorbu spermií. Pro stanovení vlivu vybraných endokrinních disruptorů na životaschopnost těchto buněk jsme používali metodu trojkombinace, založenou na kombinaci tří testů využívajících vitální barvy alamarovou modř, CFDA-AM a neutrální červeň. Zjistili jsme, že studované polyaromatické uhlovodíky, pesticidy a bisfenol A nebyly cytotoxické a ovlivňovaly viabilitu Sertoliho buněk, pouze ve vysokých koncentracích (200 µM). Ovšem antimikrobiální chemikálie přidávané do mýdel a šampónů triklokarban a triklosan, významně snižovaly metabolickou aktivitu, membránovou integritu a celkovou energetickou bilanci Sertoliho buněk. Cytotoxické působení triklokarbanu bylo potvrzeno i pomocí přístroje xCELLigence, který hodnotí viabilitu buněk v reálném čase bez nutnosti barvení buněk. Výsledky této práce potvrzují, že endokrinní disruptory mohou působit nepřímo na vývoj zralých spermií přes snížení viability, a tím i funkčnost Sertoliho buněk.

Abstract:

In recent years, professional and general public are concerned for male reproductive health and declining of its functions. There are several epidemiological studies which connect the impact of specific substances on reproductive system of humans and wildlife. The aim of this diploma thesis was to write a review about the role of Sertoli cells in the endocrine disruption and to study the effects of significant environmental pollutants on these cells that are actually very significant testicular somatic cells. Sertoli cells play a key role in formation and developing of testis and production of sperm. To determine the adverse effect of selected endocrine disruptors on these cells' viability, we used method based on combination of three tests with vital deys Alamar Blue, CFDA-AM and Neutral Red. We found out that Polyaromatic Hydrocarbons, pesticides and bisfenol A were not cytotoxic and they affect viability of Sertoli cells only at the highest concentration (200 µM). However, antimicrobial chemicals triclocarban and triclosan are added to soaps and shampoo, significantly reduced metabolical activity, membrane integrity and overall energetic balance of Sertoli cells. The cytotoxic effect of triclocarban was also confirmed by the moderm approach in cytotoxicity testing based on real-time label-free impedance monitoring. Our results of confirmed that endocrine disruptors can indirectly affect the development of mature sperm by reducing of their viability and thus the nursing function of Sertoli cells.
