Development of CRISPR/Cas9 based reporter cell models

Abstract:

Bakalářská práce se věnuje CRISPR/Cas9 systému a jeho využití v oblasti editace genomu. CRISPR/Cas9 technologie má velký potenciál pro uplatnění v rozmanitých oblastech molekulární genetiky, ve vědecké sféře je používána pro studium molekulárních mechanismů rostlin, živočichů i lidských genů, včetně pozměňování zárodečné lidské linie. CRISPR/Cas9 systém je také, ve spojení se specifickými endonukleázami, určen k editování genomu nebo k cílené regulaci genů. Editaci genomu lze provádět pomocí CRISPR systému typu II odvozeného od Streptococcus pyogenes. Onkogen Myc reguluje transkripci genů, které jsou potřebné pro koordinaci řady buněčných procesů - proliferace, růst, diferenciace, apoptóza a sebeobnovení. Nedávno bylo také prokázáno, že gen Myc slouží jako přímý regulátor biogeneze ribozomů. V této práci bylo pro zjištění a ověření poznatků z publikovaných prací exprese genu c-Myc a jejího ovlivnění RPL11 použita metoda transfekce siRNA (small interfering RNA), které se uplatňuje právě v procesu RNA interference.

Abstract:

This bachelor thesis examines with CRISPR/Cas9 system and its utilisation in genome editing. CRISPR/Cas9 technology has great potential for application in diverse areas of molecular genetics to study the molecular mechanisms of plants, animals, and human genes, including altering the germ line. CRISPR/Cas9 system is designed for genome editing or targeted gene regulation in conjunction with specifika endonucleases. Genome editing can be performed using a Streptococcus pyogenes Type II CRISPR system. Myc oncogene regulates the transcription of genes that are needed to coordinate a variety of cellular processes - proliferation, growth, differentiation, apoptosis, and self-renewal. Previous studies delineate that the Myc gene serves as a direct regulator of ribosome biogenesis. In this bachelor thesis, the method of siRNA transfection was used to determine and verify the results gene expression of Myc and its influence on RPL11.
