Metabolism of resveratrol in the colon model

Abstract:

Resveratrol je základní a zatím nejvíce zkoumaná sloučenina ze skupiny stilbenoidů. Resveratrolu i ostatním stilbenoidům je připisováno mnoho pozitivních vlastností, kterými jsou schopny působit na zdraví člověka. Resveratrol vykazuje schopnost rychlého vstřebávání a metabolismu v lidském organismu, čímž vznikají metabolity s rozdílnou chemickou strukturou. V posledních letech vzrostl zájem zejména ve zkoumání mikrobiálních metabolitů resveratrolu.Praktická část této práce byla zaměřena na zkoumání vzniku metabolitů resveratrolu pomocí bakterií, které byly přítomny ve vzorcích stolice. In vitro fermentace probíhala v 96-jamkových deep-well destičkách, ze kterých byly následně v pěti časových bodech (0,2,4,8 a 24 hodin) odebírány vzorky pro měření. Vzorky byly podrobeny NMR analýze, při které byla změřena spektra a koncentrace vzniklých metabolitů. Nakonec byly výsledky z NMR spektrometru statisticky zanalyzovány. Cílem praktické části bakalářské práce bylo stanovit hlavní katabolity resveratrolu a posoudit funkčnost nové fermentační metody.Z výsledků této práce bylo zjištěno, že chemická struktura resveratrolu prošla mikrobiální přeměnou. Hlavním a zároveň jediným metabolitem, který během fermentace vznikl, byl dihydroresveratrol. Ve vzorcích byl hledán i lunularin, který byl dříve také identifikován jako metabolit resveratrolu. Důvodem, proč byla nalezena pouze jedna nově vzniklá sloučenina, je pravděpodobně rozdílné složení střevní mikrobioty našich dárců. Fermentace v 96-jamkových deep-well destičkách byla vyhodnocena jako funkční a vyhovující pro další experimenty.

Abstract:

Resveratrol is the essential and most studied compound of the stilbenoid group. Resveratrol, as well as other stilbenoids, is attributed to many positive properties that are able to affect human health. Resveratrol has the ability to be rapidly absorbed and metabolized in the human body, thereby producing metabolites with different chemical structures. In recent years, there has been a growing interest in the study of microbial metabolites of resveratrol.The practical part of this thesis was focused on the formation of resveratrol metabolites using bacteria derived from stool samples. In vitro fermentation took place in 96-deep well plates, from which the samples for measurement were taken in five time points (0, 2, 4, 8, and 24 hours). The samples were subjected to NMR analysis and the spectrum and concentration of produced metabolites were measured. Finally, the results from NMR spectrometer were statistically analysed. The aim of the practical part of this bachelor thesis was to determine the main catabolites of resveratrol and to assess the functionality of the new fermentation method.The results of this thesis show that the chemical structure of resveratrol went through microbial transformation. The main and only metabolite, that arose during fermentation was dihydroresveratrol. Lunularin, another known metabolite of resveratrol, was also searched. The reason why the only one newly formed compound was found is probably in the different composition of the gut microbiota of our donors. Fermentation in 96-deep well plates was evaluated as functional and suitable for further experiments.
