Plant response to combined stress caused by low temperature and inadequate mineral nutrition

Abstract:

V této práci byl studován vliv abiotických stresorů, konkrétně nízké teploty a nedostatečné výživy dusíkem, a jejich vzájemné kombinace na klíčení semen. Pro porovnání byla použita semena lilku rajčete (Solanum lycopericum L.), rostliny obecně citlivé k nízkým teplotám a kedlubny (Brassica oleracea var. gongylodes), rostliny k tomuto stresovému faktoru méně citlivé. Pro simulaci nízké teploty byla použita teplota 8 °C, oproti normální teplotě 20 °C, a pro výživu byl použit Knopův živný roztok, při deficitní výživě dusíkem nebyly do tohoto živného roztoku přidány ionty dusíku. Semena byla vystavena působení uvedených faktorů po dobu 72 hodin, kdy byl sledován jejich vliv na klíčení semen. Tento experiment ukázal, že stanovená nízká teplota prodlužuje dobu potřebnou k vyklíčení semen. Tento efekt se projevuje u semen lilku rajčete daleko výrazněji než u semen kedlubny. Deficience dusíku měla v tomto případě pozitivní vliv na rychlost klíčení semen obou rostlinných druhů. Naopak semena kultivována v živném roztoku s optimální výživou vykazovala inhibici klíčení.

Abstract:

In this study, effect of abiotic stress, namely low temperature and nitrogen deficiency as well as their combined effects on seed germination, was studied. Tomato (Solanum lycopersicum L.) seeds were used for comparison as they are generally sensitive to low temperatures, and kohlrabi (Brassica oleracea var. gongylodes) seeds were used as they are less sensitive to this stress factor. To simulate low temperatures, a temperature 8 °C was used, in comparison to the normal temperature 20 °C, and for nutrition Knop nutrient solution was used, when for nitrogen deficiency solution nitrogen ions were not added to this solution. The seeds were exposed to previously mentioned factors for 72 hours, during which their effect on seed germination was monitored. Results of this experiment showed that the low temperature prolongs the time required for seed germination. This effect is much more significant for tomato seeds than for kohlrabi seeds. In this case nitrogen deficiency had a positive effect on the germination rate of seeds of both plants. On the other hand, seeds cultivated in nutrient solution with optimal nutrition showed inhibition of germination.
