Diverzita vejcorodých monogeneí (Dactylogyridae) parazitujících na afrických tetrách (Characiformes: Alestidae): strukturální a molekulární fylogenetická analýza

Diversity of oviparous monogeneans (Dactylogyridae) parasitizing African tetras (Characiformes: Alestidae): structural and molecular phylogenetic analysis

Abstract:

V rámci Afriky bylo dosud popsáno přes 240 druhů ryb řádu Characiformes spadajících do 4 čeledí: Alestidae (120 druhů), Citharinidiae (8 druhů), Distichodontidae (108 druhů) a Hepsetidae (6 druhů). Na rybách tohoto řádu bylo dosud zaznamenáno 66 druhů monogeneí čeledě Dactylogyridae náležejících do 4 rodů: Afrocleidodiscus (3 druhy), Annulotrema (48 druhů), Characidotrema (13 druhů) a Nanotrema (2 druhy). Na zástupcích čeledě Alestidae (afrotetry) bylo nalezeno 55 druhů monogeneí: Afrocleidodiscus (1 druh), Annulotrema (41 druhů) a Characidotrema (13 druhů). Předložená dizertační práce se zaměřuje na studium druhové rozmanitosti a fylogenetických vztahů monogeneí parazitujících na afrických rybách řádu Characiformes (zejména čeledi Alestidae) s použitím dvou metodologických přístupů, a to morfologické a molekulární analýzy. V průběhu let 2012 – 2018 byl zpracován parazitologický materiál z 20 druhů ryb (345 jedinců) a 8 afrických států: Demokratická republika Kongo, Jihoafrická republika, Kamerun, Keňa, Nigérie, Senegal, Súdán a Zimbabwe. Výskyt monogeneí se potvrdil u 16 druhů hostitelů (Alestidae: 12 druhů, Citharinidae: 1 druh, Distichodontidae: 2 druhy a Hepsetidae: 1 druh). Celkově bylo nalezeno 48 druhů monogeneí čeledě Dactylogyridae: 3 druhy rodu Afrocleidodiscus, 35 druhů rodu Annulotrema, 8 druhů rodu Characidotrema a 2 druhy rodu Nanotrema. Výsledkem této práce je 9 nově popsaných druhů: 6 druhů rodu Annulotrema (A. ansatum, A. bipatens, A. bracteatum, A. cucullatum, A. pontile, A. pseudonili) a 3 druhy rodu Characidotrema (C. auritum, C. pollex, C. vespertilio). Dále byla provedena redeskripce dvou druhů (Annulotrema elongatum, Characidotrema nursei) a poskytnuty nové morfologické údaje a geografické záznamy pro 7 dříve popsaných druhů (Annulotrema alestesnursi, A. besale, A. nili, A. pikoides, Characidotrema brevipenis, C. spinivaginus, C. zelotes). Na základě nových morfologických znaků byla rozšířena diagnóza rodu Annulotrema (přítomnost slabě sklerotizovaných podpůrných membrán připojených k destičkám haptoru) a Characidotrema (přítomnost slabě sklerotizovaných struktur připomínajících pochvu obepínající hrot ventrálních středních háčků). Studium taxonomického významu reprodukčních orgánů a skleritů haptoru odhlalilo, že nejdůležitějším znakem rozlišujícím druhy v rámci rodů Annulotrema a Characidotrema je morfologie samčího kopulačního orgánu. Rozdělení druhů rodu Characidotrema do dvou „morfoskupin“ na základě morfologických znaků samčího kopulačního orgánu bylo potvrzeno fylogenetickou analýzou. Využitím sekvencí 28S rDNA byly prezentovány první vhledy do fylogenetických vztahů mezi 28 druhy monogeneí parazitujících na afrických trnobřichých. Výsledný fylogenetický strom získaný metodami maximální věrohodnosti a Bayesovské analýzy rozdělil monogenea čeledě Dactylogyridae na dvě hlavní skupiny. Jedna skupina obsahuje druhy rodu Annulotrema, zatímco druhá zahrnuje dva druhy rodu Annulotrema z hostitele Hepsetus odoe (Hepsetidae) spolu s druhy zbývajících rodů (Afrocleidodiscus, Characidotrema a Nanotrema) seskupenými dle hostitelských preferencí na úrovni čeledi. Rozdělení druhů rodu Annulotrema do dvou linií naznačuje polyfyletický status tohoto rodu. Dva druhy rodu Annulotrema zaznamenané na hostiteli Hepsetus odoe (A. biaense, A. hepseti) pravděpodobně představují další jedinečnou linii a měly by být považovány za incertae sedis. Zástupci monogeneí parazitujících na afrotetrách netvořili monofyletickou skupinu, místo toho byli rozděleni do dvou nezávislých linií. První linie obsahovala druhy rodu Annulotrema a druhá zahrnovala druhy rodu Characidotrema.

Abstract:

African freshwaters harbour more than 240 characiform species currently arrayed in four families: Alestidae (120 spp.), Citharinidae (8 spp.), Distichodontidae (108 spp.), and Hepsetidae (6 spp.). To date, 66 nominal species attributed to four genera of dactylogyrid monogeneans have been reported from African Characiformes: Afrocleidodiscus (3 spp.), Annulotrema (48 spp.), Characidotrema (13 spp.) and Nanotrema (2 spp.). Alestidae (African tetras) are known to be parasitized by 55 species of three genera: Afrocleidodiscus (1 sp.), Annulotrema (41 spp.) and Characidotrema (13 spp.). The presented dissertation focuses on the study of diversity and phylogenetic relationships of monogeneans parasitizing African characiform fishes, especially those of the Alestidae, using morphological and molecular analyses. During 2012 – 2018, parasitological material was processed from 20 fish species (345 individuals) and eight African countries: the Democratic Republic of the Congo, South Africa, Cameroon, Kenya, Nigeria, Senegal, Sudan, and Zimbabwe. The occurrence of monogeneans was confirmed in 16 species of hosts representing four African families (Alestidae 12 spp., Citharinidae 1 sp., Distichodontidae 2 spp., and Hepsetidae 1 sp.). A total of 48 species of dactylogyrids were recognized: Afrocleidodiscus (3 sp.), Annulotrema (35 spp.), Characidotrema (8 spp.), and Nanotrema (2 spp.). As a result, nine dactylogyrid species were described as new to science: six species of Annulotrema (A. ansatum, A. bipatens, A. bracteatum, A. cucullatum, A. pontile, A. pseudonili) and three species of Characidotrema (C. auritum, C. pollex, C. vespertilio) Two species of monogeneans were redescribed (Annulotrema elongatum, Characidotrema nursei), and new morphological data and geographical records for seven known species were provided (Annulotrema alestesnursi, A. besale, A. nili, A. pikoides, Characidotrema brevipenis, C. spinivaginus, C. zelotes). New morphological features supplementing the diagnosis of Annulotrema (i.e. presence of lightly sclerotized supporting membranes associated with the haptoral bars) and Characidotrema (i.e. presence of a sheath-like structure relating to the ventral anchor) were revealed. Investigation of the relative taxonomic importance of the characters of the reproductive organs and those of the haptoral sclerites in accordance with molecular data revealed, that the most important feature for species differentiation within Annulotrema and Characidotrema appears to be the morphology of the male copulatory organ (MCO). The division of Characidotrema spp. into two morphogroups based on morphological features of the MCO were also supported by the results of phylogenetic analysis. Using 28S rDNA sequences, the first insights into the phylogenetic relationships among 28 species of monogeneans parasitizing African characiform fishes were presented. The resulting phylogenetic tree obtained by maximum likelihood and Bayesian analysis depicts two main groups of dactylogyrids parasitizing African characiforms: one contains Annulotrema species infecting alestids, whereas the other contains two species of Annulotrema infecting Hepsetus odoe (Hepsetidae) and species of the remaining genera (Afrocleidodiscus, Nanotrema, and Characidotrema) grouped by their host taxonomic affinity. The division of the analyzed Annulotrema species into two lineages suggests a polyphyletic status of the genus. Two species of Annulotrema described from hepsetids (A. biaense, A. hepseti) likely represent another unique lineage and they should be considered incertae sedis. Dactylogyrids parasitizing African tetras did not form a monophyletic group, instead they were found in two independent lineages. The first lineage contains species of Annulotrema, and the second includes species of Characidotrema.
