Characterization of mesenchymal stem cells from the bone marrow of broiler chickens

Abstract:

Cílem disertační práce byla optimalizace izolace, kultivace a kryokonzervace kmenových buněk izolovaných z kostní dřeně brojlerových kuřat pro účely genové banky. Další cíle disertační práce zahrnovaly hodnocení morfologie a životaschopnosti buněk, detekce markerů charakteristických pro mezenchymální kmenové buňky, schopnost diferenciace v jiné typy buněk a hodnocení ultrastruktury buněk. V disertační práci byly použity různé metody, jako jsou například průtoková cytometrie, fluorescenční mikroskopie, transmisní elektronová mikroskopie a PCR analýza. Výsledky práce ukázaly, že izolace a optimalizace kultivace byla úspěšná a podařilo se tedy izolovat mezenchymální kmenové buňky, což následně bylo ověřeno několika metodami. Po vysetí buněk byla pozorována morfologie, kdy buňky vypadaly z morfologického hlediska nejprve jako buňky kulatého tvaru, které postupně přilnuly ke dnu kultivační láhve. Postupně buňky dosáhly vřetenovitého tvaru a rychle se množily. Po 6 – 7 dnech buňky vytvořily homogenní monovrstvu buněk, která se podobala fibroblastům. Pro ověření MSC byla provedena fenotypová analýza pomocí markerů, která potvrdila přítomnost MSC CD73, CD90, CD105 a naopak ve všech pasážích vyloučila přítomnost hematopoetických markerů CD34 a CD45. Metodou RT-PCR byly dále markery potvrzeny. Pomocí histologického barvení byla zjišťována schopnost diferenciace mezenchymálních kmenových buněk na chondrocyty, osteocyty a adipocyty. Jako poslední byla sledována viabilita kmenových buněk čerstvých a následně rozmražených pro možnost dlouhodobého uchovávání buněk pro další experimenty.

Abstract:

The aim of the dissertation was to optimize the isolation, cultivation and cryopreservation of stem cells isolated from the bone marrow of broiler chickens for the purposes of the gene bank. Other objectives of the dissertation included the evaluation of cell morphology and viability, the detection of markers characteristic of mesenchymal stem cells, the ability to differentiate into other cell types, and the evaluation of cell ultrastructure. Various methods were used in the dissertation, such as flow cytometry, fluorescence microscopy, transmission electron microscopy, and PCR analysis. The results of the work showed that the isolation and optimization of the cultivation was successful and it was possible to isolate mesenchymal stem cells, which was subsequently verified by several methods. After seeding of the cells, the morphology was observed, when the cells first appeared from a morphological point of view as round-shaped cells, which gradually adhered to the bottom of the culture flask. Gradually the cells reached a spindle shape and multiplied rapidly. After 6-7 days, the cells formed a homogeneous monolayer of cells that resembled fibroblasts. To verify the MSC, a phenotypic analysis using markers was performed, which confirmed the presence of MSC CD73, CD90, CD105 and, on the contrary, ruled out the presence of hematopoietic markers CD34 and CD45 in all passages. The markers were further confirmed by the RT-PCR method. Using histological staining, the ability of mesenchymal stem cells to differentiate into chondrocytes, osteocytes and adipocytes was determined. Lastly, the viability of fresh and subsequently thawed stem cells was monitored for the possibility of long-term storage of cells for further experiments.
