Abstract:

Tématem této diplomové práce je Příprava karbamátů R-1-(6-fluorbenzthiazol-2-yl)-3-(methylthio)propan-1-aminu a stanovení jejich inhibičních aktivit vůči enzymům. Teoretická část je zaměřena na znalosti inhibitorů cholinesteráz, konkrétně karbamáty a benzthiazoly, jako jsou jejich struktura, syntéza či funkce v organismu. Dále se teoretická část práce věnuje obecnému charakteru cholinesteráz, jejich struktuře a biologické funkci. V další části je poté sepsáno několik obecných znalostí o inhibitorech cholinesteráz, o typech inhibice a jejich principech. Experimentální část diplomové práce poté zahrnuje 2 části - syntetickou a biologickou. Syntetická část spočívá v přípravě a syntéze celkem 10 karbamátů. Kapitola obsahuje postupy a schémata přípravy výchozí látky i syntéz jednotlivých karbamátů. Dále obsahuje jejich hodnoty z NMR spektrometru spolu s bodem tání. Biologická část se zaměřuje na měření inhibičních vlastností karbamátů vůči cholinesterázám (acetylcholinesteráze a butyrylcholinesteráze). Konkrétně se zde stanovuje enzymatická aktivita cholinesteráz, IC50 neboli 50% inhibiční koncentrace a kinetické parametry, na jejichž základě lze určit typ inhibice. Na závěr je uvedena kapitola se všemi výsledky společně s diskuzí.

Abstract:

The topic of this thesis is the preparation of R-1-(6-fluorobenzthiazol-2-yl)-3-(methylthio)propan-1-amine carbamates and the determination of their inhibitory activities against enzymes. The theoretical part is focused on knowledge of cholinesterase inhibitors, specifically carbamates and benzthiazoles, such as their structure, synthesis and function in the body. Furthermore, the theoretical part of the work is devoted to the general character of cholinesterases, their structure and biological function. In the following part, some general knowledge about cholinesterase inhibitors, types of inhibition and their principles is written out. The experimental part of the thesis includes two parts - synthetic and biological. The synthetic part consists in the preparation and synthesis of a total of 10 carbamates. The chapter contains procedures and schemes for the preparation of the starting material and the synthesis of individual carbamates. It also contains their values from the NMR spectrometer along with the melting point. The biological part focuses on measuring of inhibitory properties of carbamates against cholinesterases (acetylcholinesterase and butyrylcholinesterase). Specifically, cholinesterase enzymatic activity, IC50 or 50% inhibitory concentration and kinetic parameters are determined here, on the basis of which the type of inhibition can be determined. At the end, there is a chapter with all the results together with a discussion.
