Abstract:

Petunia hybrida Vilm. je jednou z nejdůležitějších letniček. Tržní niku v sortimentu představuje skupina tetraploidních odrůd, které jsou zajímavé díky svému obřímu květu. Jedná se však pouze o staré OP (open pollinated) odrůdy. Vytvoření homozygotních tetraploidních linií je předpokladem pro výrobu odrůd F1. Za tímto účelem jsme použili 9 homozygotních linií používaných v komerčních F1 hybridech. Týden staré semenáče pěstované na MS médiu s agarem byly ošetřeny roztokem 30 μM oryzalinu po dobu 24 hodin v podmínkách in vitro. Později byly pěstovány ve skleníku. Cytofotometricky byl určen stupeň ploidie. Výtěžek tetraploidů byl 20–70 %. Byly získány rostliny s genotypem GGGG a gggg, včetně rostlin s červeným květem. Velikost květu se zvýšila u tetraploidů průměrně o 30 %. F1 kříženci, kteří vznikli z dvojice indukovaných tetraploidů, byli uniformní, s velkým květem, některé v typu starých OP odrůd var. superbissima. Byl vytvořen polyploidizační laboratorní protokol a návrh uceleného šlechtitelského programu ke šlechtění F1 polyploidních odrůd.

Abstract:

Petunia hybrida Vilm. is one of the most important annuals. The market niche in the assortment is represented by a group of tetraploid varieties, which are interesting due to their giant flower. However, these are only old OP (open pollinated) varieties. The formation of homozygous tetraploid lines is a prerequisite to produce F1 varieties. For this purpose, we used 9 homozygous lines used in commercial F1 hybrids. Week-old seedlings grown on MS agar medium were treated with a solution of 30 μM oryzalin for 24 hours under in vitro conditions. They were later grown in a greenhouse. The degree of ploidy was determined cytophotometrically. The yield of tetraploids was 20-70%. Plants with the GGGG and gggg genotypes were obtained, including plants with a red flower. Flower size increased by an average of 30% for tetraploids. F1 hybrids that originated from a pair of induced tetraploids were uniform, with a large flower some in the type of old OP varieties var. superbissima. A polyploidization laboratory protocol and a design of a comprehensive breeding program for the breeding of F1 polyploid varieties were created.
