The proliferation of undifferentiated human embryonic stem cells in chemically defined culture media

Abstract:

Lidské embryonální kmenové buňky (lidské ES buňky) jsou díky své neomezené proliferační kapacitě a možnosti diferencovat v jakýkoliv buněčný typ lidského těla jedinečným zdrojem pro cílené buněčné terapie. Současný kultivační systém, využívající podkladové výživné vrstvy fibroblastů a médium s obsahem potenciálně patogenních a imunogenních látek, však toto prozatím znemožňuje. Chemicky definovaná média, která se podařilo vytvořit v několika posledních letech, vyžadují vysoké koncentrace růstových faktorů a extrémně tak zvyšují cenu média. V běžné kultivaci je proto není možno rutinně využívat. Cílem této práce bylo testování rostlinného produktu VegetaCell pro kultivaci lidských ES buněk a vytvoření nového kultivačního média za pomocí dalších, chemicky definovaných přídavků. Lidské ES buňky byly ve standardním médiu adaptovány na kultivaci za pomocí lidských předkožkových fibroblastů. Z média pak byl odstraněn KnockOut Serum Replacement, byl nahrazen VegetaCell a doplněn dalšími chemicky definovanými přídavky bez zvířecích proteinů. Médium nebylo potřeba doplnit žádnými růstovými faktory. V tomto novém médiu lidské ES buňky proliferovaly porovnatelně s buňkami ve standardním médiu. Hladina apoptózy byla v médiích s VegetaCell snížena. Kultivované buňky exprimovaly všechny pozorované markery nediferencovaných lidských ES buněk.

Abstract:

The unlimited proliferation capacity and ability to differentiate into all cell types of human body are the greatest promises of human embryonic stem cells (human ES cells). The current culture system, based on fibroblast feeder layers and medium containing potentially pathogenic and immunogenic components, limits the usage of human ES cells in regenerative medicine. In last several years, numerous chemically defined media were established. They require high doses of several growth factors, making them impossible to use in routine cell cultures. The aim of this study was to test the plant product called VegetaCell on cultured human ES cells and to create a new culture medium with other, chemically defined supplements. I adapted human ES cells culture on human foreskin fibroblasts in the standard medium. Then I deprived the medium of KnockOut Serum Replacement, replaced it by VegetaCell and replenished with other chemically defined and xeno-free supplements. No additional growth factors were needed. In this new optimized medium, human ES cells were able to proliferate in the comparable level with cells in the standard medium. The apoptosis level was reduced when compared with cells in the standard medium on human foreskin feeder layer. They also expressed all pursued markers of undifferentiated human ES cells.
