Detection of the toxic effects of synthetic dyes by determining the inhibition of growth of Lemna minor

Abstract:

Předložená diplomová práce se zaměřuje na možnost využití inhibice růstu Lemna minor pro hodnocení toxicity syntetických barviv a vzorků odpadních vod. Cílem práce bylo kvantifikovat toxické účinky syntetických barviv Methylene Blue a Reactive Orange 16 a dvou vzorků odpadních vod z textilního závodu na vegetativní růst okřehku menšího (Lemna minor), během biodegradace pomocí lignolytické houby Irpex lacteus. Hodnotícím kritériem byla inhibice růstu počítaná ze dvou parametrů, z množství biomasy a růstové rychlosti. Hlavním výstupním ukazatelem byla hodnota EC50. Jako referenční látky byly použity 3,5-dichlorfenol a dichroman draselný. Biodegradace byla účinná u všech sledovaných látek. Barvivo Methylene Blue i vzorky odpadních vod z textilního závodu byly nejvíce toxické na vstupu, tedy před zahájením biodegradace. U biodegradačních meziproduktů i v koncových produktech byl prokázán pokles toxických účinků. V případě, že byly inhibiční účinky zjištěny, byla vypočtena hodnota EC50. Koncentrace chlorofylu a a feopigmentů byly stanoveny u obou referenčních látek a u dvou produktů reálných vzorků - meziprodukt odpadní vody I a výstupního vzorku odpadní vody II.

Abstract:

The present diplom thesis focuses on the potential for growth inhibition of Lemna minor to evaluate the toxicity of synthetic dyes and wastewater samples. The aim was to quantify the toxic effects of synthetic dyes Methylene Blue and Reactive Orange 16 and samples of wastewater from textile plant for vegetative growth of duckweed (Lemna minor) during biodegradation by fungi lignolytické Irpex lacteus. The evaluation criterion was the inhibition of growth calculated from two parameters of biomass and growth rate. The main output indicator was EC50. As reference substances were used 3.5-dichlorophenol and potassium dichromate. Biodegradation was effective in all monitored substances. Dye Methylene Blue and samples of wastewater from the plant textile plant were the most toxic of the input, thus before biodegradation. The biodegradation intermediates and final products were show to decrease the toxic effects. In the event that the inhibitory effect observed was calculated EC50. The concentration of chlorophyll a and feopigments were determined for both reference materials and real samples of two products ? intermediate wastewater I and final sample wastewater II.
