Variability of histone post-translational modifications in individuals with normozoospermia

Abstract:

Spermie jsou vysoce specializované buňky, jejichž hlavní úlohou je dodat oocytu otcovský epigenom. Během spermiogeneze dochází k významné reorganizaci chromatinu, na které se podílejí hlavně histony a jejich specifické posttranslační modifikace. Tyto proteoformy patří mezi jeden z klíčových epigenetických faktorů, které ovlivňují výměnu histonů za protaminy. Změny během těchto událostí mohou zásadním způsobem ovlivnit mužskou reprodukci. Základní laboratorní vyšetření mužské plodnosti, spermiogram, nedokáže zdaleka odhalit všechny případy neplodnosti ani jejich příčinu. Pro zavedení přesnějších diagnostických testů je zapotřebí pochopit mechanismy poruch mužské plodnosti na molekulární úrovni. Cílem diplomové práce bylo charakterizovat sekvenční varianty histonů H3 a H4 spolu s jejich posttranslačními modifikacemi u pilotního souboru jedinců s normozoospermií. Analýza histonů byla provedena pomocí přístupů založených na hmotnostní spektrometrii. Proteomická data byla spolu s daty kvalitativními statisticky zpracována a na základě variability získaných výsledků byli nalezeni jedinci s odlehlými parametry histonových proteoforem. Přístupy pro charakterizaci histonů zavedené v této pilotní analýze představují základ pro následnou histonovou analýzu v statisticky významném souboru pacientských vzorků s rozmanitými poruchami mužské plodnosti.

Abstract:

Spermatozoa are highly specialized cells whose main role is to deliver the paternal epigenome to the oocyte. During spermiogenesis, significant chromatin reorganization occurs, mainly involving histones and their specific post-translational modifications. These proteoforms are one of the key epigenetic factors that influence the replacement of histones for protamines. Changes during these events can profoundly affect male reproduction. The basic laboratory test of male fertility, the spermiogram, is far from being able to detect all cases of infertility or their cause. Understanding the mechanisms of male fertility disorders at the molecular level is needed to establish more accurate diagnostic tests. The aim of this thesis was to characterize sequence variants of histones H3 and H4 along with their post-translational modifications in a pilot cohort of normozoospermic individuals. Histone analysis was performed using mass spectrometry based approaches. Proteomic data were statistically processed together with qualitative data and individuals with outlying histone parameters were identified based on the variability of the results obtained. The histone characterization approaches established in this pilot analysis provide a basis for subsequent histone analysis in a statistically significant cohort of patient samples with diverse male fertility disorders.
