Bicyclo[1.1.1]pentane Derivatives

Abstract:

Tato disertační práce se zabývá možnostmi přípravy nových derivátů bicyklo[1.1.1]pentanu (BCP) substituovaných jak v můstkových, tak v bridgehead pozicích. První část je zaměřena na syntézu BCP s cis-endo konfigurací substituentů na můstcích. Několikastupňovou syntézou byl získán výchozí derivát [1.1.1]propellanu, 9,9-dimethyl-8,10-dioxapentacyklo¬[5.3.0.02,5.03,5.03,6]dekan. Do jeho bridgehead poloh byly následně úspěšně instalovány různé funkční skupiny (SAc, COOCH3 a C≡C-R). Selektivní transformací substituentů v můstkových polohách pak byly získány reaktivní 1,2- diketony, které byly následně převedeny na stabilní deriváty chinoxalinu. Druhá část práce se zabývá přípravou rigidní trojúhelníkovité molekuly z rodiny shape-persistent makrocyklů. V několika krocích byly připraveny prekurzory budoucího makrocyklu (1,3-diethynylbicyklo¬[1.1.1]pentan a 3,6-dijod-9,10-bis(hexyloxy)fenanthren), které byly poté spojeny do finálního útvaru sérií palladiem katalyzovaných reakcí. Následné analýzy (např. MALDI-TOF, AFM) potvrdily, že trojúhelník ochotně vytváří agregáty a membrány. Jejich podrobná struktura je předmětem dalšího studia. Závěrečná část práce pojednává o syntéze různě dlouhých (0,5 – 2 nm) bifunkčních tyčinkovitých molekul odvozených od 1,3 diethynylbicyklo[1.1.1]pentanu. Do jeho koncových poloh byly instalovány různé funkční skupiny (COOH, PPh2, POPh2, p-C2B10-COOH, p-C6H4-COOH, atp.). Mnoho z těchto derivátů bylo podrobeno RTG strukturní analýze. Získané látky mohou nalézt uplatnění např. při výrobě funkčních materiálů (molekulární vodiče nebo izolátory), MOFů (MOF = Metal-Organic Framework) nebo různých zařízení na molekulární úrovni (molecular level devices).

Abstract:

The thesis deals with syntheses of a new class of bridge and bridgehead substituted bicyclo[1.1.1]pentane (BCP) derivatives. Syntheses of BCP derivatives with cis-endo configuration of bridge substituents are presented in the first part of the thesis. As a suitable starting compound was chosen a known [1.1.1]propellane derivate, 9,9-dimethyl-8,10-dioxapentacyclo[5.3.0.02,5.03,5.03,6]decane, which was obtained in a multistep procedure. Different functional groups were then successfully installed into its bridgehead positions (SAc, COOCH3 and C≡C-R). Selective transformations of the bridge substituents provided highly reactive 1,2 diketones, which were transformed into more stable quinoxaline derivatives in some cases. The second part of the thesis is focused on preparation of a rigid, triangle-like molecule, an example of a shape-persistent macrocycle. Precursors of the triangular macrocycle, 1,3-diethynyl[1.1.1]bicyclopentane and 3,6-diiodo-9,10-bis(hexyloxy)phenanthrene, were synthesized and assembled into the final system using a serie of palladium catalyzed reactions. A tendency of the molecular triangle to form aggregates and membranes was studied by various analytical methods (e.g. MALDI-TOF, AFM). The final part of the thesis deals with syntheses of rod-like molecules of different length (0,5 – 2,0 nm). The compounds are derived from 1,3 diethynylbicyclo[1.1.1]pentane with terminal positions substituted by different functional groups (e.g. COOH, PPh2, POPh2, p-C2B10-COOH, p C6H4-COOH). Many of those derivatives were characterized by X-ray diffraction. The prepared molecules promise to be useful in syntheses and/or “bottom-up” assembly of functional materials (molecular line wires or insulators), MOFs (Metal-Organic Framework) or molecular level devices.
