Function of desaturases and elongases at lipid metabolism in type 2 diabetic patients

Abstract:

Předložená diplomová práce se zabývá měřením aktivit -5, -6, a -9 desaturáz a elongáz ve všech třídách plazmatických lipidů, u insulinem léčených diabetiků typu 2 a kontrolní skupiny zdravých osob. Byla vyhodnocena závislost uvedených enzymů na obsahu glykovaného hemoglobinu. Diplomová práce obsahuje rovněž statistické vyhodnocení naměřených koncentrací mastných kyselin a dostupných biochemických dat. Se svolením Univerzity v Tübingenu bylo vyšetřeno 27 vzorků EDTA plazmy od insulinem léčených diabetiků a 20 kontrolních vzorků zdravých dárců. Pomocí preparativní chromatografie na tenké vrstvě byly tyto vzorky rozděleny do pěti frakcí (fosfolipidy, diacylglyceroly, volné mastné kyseliny, triacylglyceroly a estery cholesterolu) a přítomné mastné kyseliny byly převedeny na methylestery. Závěrečným krokem laboratorní analýzy bylo stanovení esterů mastných kyselin plynovou chromatografií. Získané výsledky byly zpracovány v programu Microsoft Office Excel 2007 a statisticky vyhodnoceny programem Statistica 12. Pomocí statistických analýz nebyly zjištěny významné změny v aktivitách desaturáz a elongázy mezi kontrolní skupinou a diabetiky, což potvrzuje účinnost léčby diabetu insulinem. Rovněž nebyly zjištěny podstatné rozdíly v zastoupení a spektru mastných kyselin v jednotlivých lipidových frakcích. Je proto možné konstatovat, že i přes zvýšenou hladinu glykovaného hemoglobinu a zvýšenou glykémii nedochází u insulinem léčených diabetiků k vážným poruchám lipidového metabolismu a k výraznému snížení aktivit lipogenních enzymů a to jak v tukové tkáni, tak i v játrech.

Abstract:

This diploma thesis evaluates the activity of -5, -6, and -9 desaturase and elongase in all classes of lipids in insuline treated diabetic patients and control group of healthy people. We processed dependency of individual monitored enzymes on the content of glycated hemoglobin. Finally, the thesis includes statistical evaluation of measured values of fatty acids and available biochemical markers. With the permission the University of Tubingen were examined 27 samples of EDTA plasma of insuline treated diabetic patients and 20 control samples from healthy donors. By preparative thin-layer chromatography, the samples were divided into five fractions (phospholipids, diglycerides, free fatty acids, triglycerides and cholesterol esters), and subsequently converted to fatty acid methyl esters. The final step was determination of fatty acid esters by gas chromatography. The results were processed in Microsoft Office Excel 2007 and statistically evaluated by program Statistica 12. Using statistical analyzes, there were no significant changes in the activities of the desaturase and elongase between the control group and diabetics, vonfirming the effectiveness of the treatment of diabetes with insulin. Also significant differences were not found in the representation of a range of fatty acids in various lipid fractions. It is therefore possible to say that despite the elevated level of glycated hemoglobin and blood glucose level does not occur at insulin treated diabetics to seriously impaired lipid metabolism and significant reduction of lipogenic enzyme activities, both in adipose tissue and in the liver.
