Identification of "\kur{Arcobacter}-like Species" by PFGE

Abstract:

Práce pojednává o \kur{Arcobacter\kur{-like species a možnostech jejich typizace, především pomocí metody pulzní gelové elektroforézy. Cílem této práce je čtenáře v první části seznámit s charakteristikou a problematikou výskytu bakterií \kur{Arcobacter\kur{-like species. Další část je věnována podrobnému popisu metody pulzní gelové elektroforézy a dalším metodám pro typizace mikroorganismů. V rámci této diplomové práce byla v první řadě metoda PFGE optimalizována a zavedena pro typizaci vybraných sbírkových kmenů a izolátů \kur{Arcobacter\kur{-like species. Dále byla provedena izolace kmenů \kur{Arcobacter\kur{-like species pro ověření kmenové diverzity v rámci vzorků drůbežího masa s následnou identifikací těchto izolovaných kmenů pomocí mPCR.

Abstract:

This work deals with \kur{Arcobacter\kur{-like species and the possibilities of their typing, especially using the method of pulse-field gel electrophoresis. The aim of this work is to get acquaint the reader with the characteristics and problems of the occurrence of bacteria \kur{Arcobacter\kur{-like species in the first part. The next part is devoted to a detailed description of the method of pulse-field gel electrophoresis and other methods for typing microorganisms. In the framework of this thesis the PFGE method was optimized and introduced for the typing of selected collection strains and isolates of \kur{Arcobacter\kur{-like species. Furthermore the isolation of \kur{Arcobacter\kur{-like species strains was performed to verify the strain diversity within the poultry meat samples. It was followed by the identification of these isolated strains by mPCR.
