Abstract:
Úvod. Somatické mutace v genech BRAF a NRAS doprovázejí přibližně 45% a 17% melanomů. Tyto mutace vedou ke konstitutivní aktivaci signální dráhy MEK-ERK. Častý výskyt mutace BRAFV600E vedl k vývoji léčiv cílených na BRAF, která účinně blokovala signalizaci MEK-ERK v melanomech s mutací BRAF. Background. Somatic mutations in BRAF and NRAS genes are found in about 45% and 17% of melanomas respectively. These mutations lead to a constitutive activation of the MEK-ERK signaling pathway. The frequent occurrence of the BRAFV600E mutation led to the development of BRAF-targeted drugs which effectively block the MEK-ERK signaling in BRAF-mutant melanoma cells.
