Escape reactions of Drosophila melanogaster larvae induced by the presence of entomopathogenic nematodes and their products

Abstract:

Octomilka Drosophila melanogaster je drobný hmyzí organismus, na kterém se po desítky let provádějí nejrůznější druhy experimentů. Nejen dospělci, ale i larvy octomilek mají velmi dobře popsaný smyslový aparát a detailně prozkoumanou nervovou soustavu. Tato práce shrnuje základní poznatky o smyslovém ústrojí ve vztahu k reakcím na vnější podněty nejrůznějšího charakteru. Díky relativně propracované smyslové soustavě dokážou octomilky na stimuly z okolí adekvátně odpovědět. Na pozitivní podněty octomilka reaguje atrakcí, na negativní naopak únikovým chováním a specifickými pohyby. Souhrnně se jedná o aspekty behaviorální imunity. Pro studium behaviorální imunity larev Drosophila melanogaster byly zvoleny entomopatogenní hlístice Heterorhabditis bacteriophora. Jde o patogenní druh hlístic, který pomocí řady toxických látek parazituje na hmyzím hostitelském organismu. Cílem experimentální části této práce je ověřit hypotézu, zda larvy octomilek mění svoje chování v přítomnosti entomopatogenních hlístic. Sledování interakcí larev a hlístic umožnila metoda FIMTrack. Jde o moderní technologii, která pomocí infračerveného světla dokáže za vysokého rozlišení sledovat velké množství larev a vyhodnocovat široké spektrum parametrů. Ačkoliv se nepodařilo ověřit únikové chování larev Drosophila melanogaster, metoda FIMTrack přesto zprostředkovává jedinečný náhled na interakci patogenů a jejich hostitele. Právě detailní průzkum těchto vazeb by mohl mít v budoucnu potenciál nejen ve vědeckém výzkumu, ale i hospodářské sféře.

Abstract:

The fruit fly Drosophila melanogaster is a small art of insect organism on which various kinds of experiments have been carried out for decades. Not only adults, but also larvae have a very well described sensory apparatus and nervous system. This work summarizes the basic knowledge of the sensory system in relation to reactions to external stimuli. Thanks to a relatively sophisticated sensory system, fruit flies can respond adequately to stimuli from their surroundings. The fruit fly responds to positive stimuli with attraction movements in contrast to negative ones, where escape behavior is used. These are aspects of behavioral immunity. To observe the behavioral immunity of fruit fly larvae, the pathogenic species of nematodes Heterorhabditis bacteriophora were chosen. These entomopathogenic nematodes, with the help of a number of toxic substances, parasitizes the insect host organism. The main aim of the experimental part of this work is to verify the hypothesis of whether fruit fly larvae change their behavior induced by the presence of entomopathogenic nematodes. Monitoring the interaction of larvae and entomopathogenic nematodes was enabled by the FIMTrack method. It is a modern technology that uses infrared light to monitor a large number of larvae in high resolution and evaluate a wide range of parameters. Although the escape behavior of Drosophila melanogaster larvae has not been verified, the FIMTrack method provides a unique insight into the interaction of pathogens and their hosts. A detailed exploration of these links could have potential in the future not only in scientific research but also in the economic sphere.
