Determination of biomass and viability in stressed grapevine cell suspension cultures

Abstract:

Vinná réva patří mezi významné hospodářské rostliny, a proto je důležité co nejlépe porozumět její schopnosti reagovat na nepříznivé podmínky biotického či abiotického původu. Trendem posledních let je nahrazování zcela syntetických pesticidů a postřikových látek různými alternativami. Jedním z možných řešení je využití vlastního aparátu rostliny pro včasné aktivování mechanismů rezistence vůči nepříznivým vlivům. Obsahem této práce je shrnutí dosavadních poznatků o odpovědi na stresové faktory a zjišťování vhodné intenzity těchto faktorů pro další studium. Byla sledována životaschopnost buněčných kultur révy vinné a nárůst biomasy těchto kultur po působení různých typů stresu. Výsledky ukazují, že různě staré buňky vykazují vůči stejným stresorům podobné reakce, liší se pouze jejich intenzita. Jediný biotický stres, houba Botrytis cinerea, se v daném časovém úseku ukázal být z hlediska životnosti buněk celkem neškodný. Z abiotických stresů byl nejméně účinný stres mechanický, který na životaschopnost ani celkovou populaci neměl téměř žádný vliv. Z teplotních faktorů se 35°C ukázalo být spíše stimulující okolností, podporující životaschopnost i nárust biomasy, naopak 10°C působilo zejména na pětidenní buňky, přičemž se velmi významně snížila jak životaschopnost, tak i celkový počet buněk. Jako nejsilnější se ukázal salinitní stres, přičemž v krátkém časovém intervalu jsou mladší (třídenní) buňky schopny snášet různou výši koncentrace NaCl lépe, než buňky starší.

Abstract:

Grapevine is ranked among important economic plants, therefore it is important to understand its ability to respond to adverse conditions of biotic or abiotic origin as much as possible. The trend in recent years is to entirely replace synthetic pesticides and spraying materials with various alternatives. One possible solution is using the plant's own machinery for early activation of resistance mechanisms. The content of this thesis is to summarize current knowledge about the response to stress factors and to determine the appropriate intensity of these factors for further study. The viability and biomass growth of grapevine cell cultures after exposure to various types of stress were studied. The results show that cells of different age exhibit similar reactions to the same stress, differing only in the intensity. The only biotic stress, fungus Botrytis cinerea has proven to be harmless in terms of cell viability in the time course of the experiment. Among the abiotic stresses the mechanical was the least effective showing almost no influence on the viability or the overall population. The temperature factor of 35°C seemed to be a rather stimulating circumstance, supporting viability and biomass growth , while 10°C influenced mainly five-day cells, reducing both the viability and the total number of cells very significantly. The salt stress showed to be the strongest, wherein during the short time interval the younger (three days) cells are able to tolerate different amounts of NaCl concentration better than older cells.
