Identifikace genů exprimovaných leukocyty vakcinovaných a nevakcinovaných kuřat po infekci Salmonella enterica

Identification of genes expressed by chicken leukocytes from vaccinated and naive chickens infected with Salmonella enterica

Abstract:

Imunitní odpověď kuřat na infekci Salmonella enterica je charakterizována nejčastěji na úrovni celých tkání sleziny nebo slepého střeva. Jednotlivé leukocytární subpopulace se však můžou podílet na této komplexní reakci různě a jejich podrobnější charakterizace chybí. V této práci jsme se proto zaměřili na charakterizaci odpovědi na infekci salmonelou u vybraných leukocytárních subpopulací ve slezině kuřat. Proteinová exprese byla charakterizována u leukocytů ze tří experimentálních skupin kuřat, a to neinfikované, nevakcinované a infikované, a vakcinované a infikované. Pomocí průtokové cytometrie byly ze sleziny sortovány subpopulace makrofágů, heterofilů, CD4+ T-lymfocytů, CD8+ T-lymfocytů, γδ T-lymfocytů a B-lymfocytů. Proteom sortovaných subpopulací byl následně charakterizován a kvantifikován pomocí hmotnostní spektrometrie. V první části práce byl nejprve charakterizován proteom všech studovaných vzorků a subpopulace byly dále analyzovány v rámci tří skupin, skupina T-lymfocytů (CD4+ T-lymfocyty, CD8+ T-lymfocyty, γδ T-lymfocyty), skupina fagocytujících leukocytů (makrofágy, heterofily) a samostatná subpopulace B-lymfocytů. Proteom CD8+ T-lymfocytů byl obohacen o proteiny buněčné diferenciace. γδ T-lymfocyty ve zvýšené míře exprimovaly malé GTPázy typu Rab. Proteom CD4+ T-lymfocytů byl obohacen o proteiny s ubiquitin SUMO-like a BAR doménami. Po infekci salmonelou došlo u T-lymfocytů ke změně v expresi celkem u 171 proteinů. U všech subpopulací T-lymfocytů na infekci odpovídaly zvýšenou expresí proteiny ANXA2, MGST1, MRP126, TXNDC5, LECT2, Gga.31975 (necharakterizovaný transportní protein vázající železo), SERPINB10 a HSP90B1. U všech T-lymfocytů byla po infekci shodně detekována snížená exprese albuminu. Proteom makrofágů byl charakteristický expresí receptorových proteinů a proteinů fagocytózy nebo prezentace antigenu. Pro proteom heterofilů byly charakteristické antibakteriální proteiny a inhibitory serinových proteáz. Makrofágy na infekci salmonelou odpovídaly zvýšenou expresí proteinů MRP126, CATHL1, CATHL2, GAL1, CTSB, CTSC, RSFR, SOD1, LECT2, LY86 a FTH. U heterofilů byla po infekci detekována snížená exprese proteinů LYZ, LYG2, CTSG, CTSL1, CATHL1, CATHL2, RSFR, MMP9 a LECT2. U B-lymfocytů byla po infekci salmonelou detekována zvýšená exprese proteinů PCNA, Ex-FABP, YWHAB, CACYBP, BTF3, LDHB, CCT7 a snížená exprese u proteinů HEXB, NPC2, SCIN, SH3BGRL3, CKB, TGM4, CTSD, CAST a PASCIN3. V druhé části práce jsme díky paralelní izolaci proteinů a RNA ověřili změny v expresi i na úrovni mRNA pomocí metody kvantitativní real-time PCR. Ve většině případů se výsledky získané pomocí proteinové hmotnostní spektrometrie potvrdily také na úrovni RNA.

Abstract:

The immune response of chickens to Salmonella enterica infection is mostly characterized in total splenic or caecal tissues. Individual leukocytic subpopulations can, however, take part in this complex reaction differently and, unfortunately, a detailed characterization of their response to Salmonella infection is missing. The aim of this study was to characterize the response of selected leukocyte subpopulations from chicken splenic tissue to intravenous infection with Salmonella Enteritidis. Leukocytes were isolated from three experimental groups of chicken: non-infected, non-vaccinated and infected, vaccinated and infected. Subpopulations of macrophages, heterophils, CD4+ T-lymphocytes, CD8+ T-lymphocytes, γδ T-lymphocytes and B-lymphocytes were sorted from the spleen by flow cytometry and their proteome was characterized and quantified by mass spectrometry. In the first part of this study the proteome of all studied samples was characterized and subpopulations were further analyzed in 3 groups: T-lymphocytes (CD4+ T-lymphocyte, CD8+ T-lymphocyte, γδ T-lymphocyte), phagocytic leukocyte (macrophages, heterophils) and a separate subpopulation of B-lymphocytes. Proteome of the CD8+ T-lymphocyte subpopulation was enriched with cell differentiation proteins. Proteome of γδ T-lymfocyte subpopulation was enriched with small Rab GTPase family proteins. Proteom of the CD4+ T-lymphocyte subpopulation was enriched with proteins with ubiquitin SUMO-like and BAR domains. After Salmonella Enteritidis infection, 171 proteins changed expression in all T-lymphocytes subpopulations. All T-lymphocyte subpopulations showed an increased expression of ANXA2, MGST1, MRP126, TXNDC5, LECT2, Gga.31975 (uncharacterized transport protein binding iron), SERPINB10 and HSP90B1 proteins after salmonella infection. A downregulated expression in all T-lymophocytes was recorded only for albumin ALB protein. Proteome of macrophages was characterized by receptor proteins, phagocytic proteins or proteins of antigen presentation. Proteome of heterophils was characterized by antibacterial proteins or serine protease inhibitors. Macrophages reacted to infection with upregulated expression of proteins MRP126, CATHL1, CATHL2, GAL1, CTSB, CTSC, RSFR, SOD1, LECT2, LY86 and FTH. Heterophiles reacted to infection with downregulated expression of proteins LYZ, LYG2, CTSG, CTSL1, CATHL1, CATHL2, RSFR, MMP9 and LECT2. The B-lymphocyte subpopulation responded to infection with upregulated expression of proteins PCNA, Ex-FABP, YWHAB, CACYBP, BTF3, LDHB, CCT7 and downregulated expression of proteins HEXB, NPC2, SCIN, SH3BGRL3, CKB, TGM4, CTSD, CAST and PASCIN3. In the second part of this study, the expression was verified at the level of RNA by quantitative real-time PCR. In most cases, the results obtained by mass spectrometry were also confirmed at the RNA level.
