Abstract:

Předložená práce se zabývá metodikami fixace a zpracování parazitické skupiny Monogenea. Monogenea jsou běžnou součástí společenstev parazitů kostnatých ryb ve všech biotopech a mohou napadat i některé obojživelníky, plazy, hlavonožce, korýše. Většina zástupců má volně plovoucí stádium larvy – onkomiracidium, která přímo napadá konečného hostitele (Kearn, 1994). Často se s nimi setkáváme také v akvakulturách a akvaristice, kde mohou způsobit značné ztráty (Ergens, 1983). Existuje mnoho metodických postupů užívaných při fixaci, zpracování a studiu monogeneí. Práce se blíže věnuje metodice převádění dočasných mikroskopických preparátů monogeneí rodu Dactylogyrus v glycerin amonium pikrátu (GAP) na trvalé preparáty v kanadském balzámu a jejich archivaci. Součástí archivace je měření, fotodokumentace a kreslení. Dočasné preparáty vydrží čitelné pouze několik let. Mnoho preparátů různých druhů monogeneí z různých hostitelů získaných zejména z ekologických studií za posledních několik let postupně degradují. Důvodem je, že GAP začne po pár letech krystalizovat, preparát vysychá a determinační znaky přestávají být viditelné. Naproti tomu je kanadský balzám trvalý. Archivované preparáty budou v budoucnu použitelné jako srovnávací materiál. Poslední část práce se zabývá vytvořením databáze. Tato databáze se později stává podkladem pro tvorbu checklistu monogeneí sladkovodních ryb sledovaných ve vybraných lokalitách v povodí řek České republiky.

Abstract:

This work deals with methods of fixing and processing of parasitic group of Monogenean. Monogeneans are a common part of communities of parasitic bony fish in all habitats. They can attack some amphibians, reptiles, cephalopods, crustaceans. The most of deputies have free floating stage of larvae – oncomiracidium which attacts directly the final host (Kearn, 1994). They are often seen in the aquacultures and the aquaristic where they can cause considerable losses (Ergens, 1983). There exist many methods used in fixation, processing and studying of monogeneans. The work specifies the method of the transfer from temporary microscopic preparations of monogenean genus Dactylogyrus in glycerine ammonium picrate (GAP) into permanent ones in canada balsam and their archiving. Parts of archiving are measurement, photos and drawing. Temporary preparations are readable only for a few years. Many various types of monogenean preparations from various hosts gained from ecological studies have gradually degraded within few last years. The reason is that the GAP starts crystallizing after a few years and the preparation dries up and determination characters stop to be visible. However the canadian balsam is permanent. Archived preparations will be used as a comparative material in the future. The final part of the work deals with the creation of a database. This database will become later the basic for the creation of monogenean checklist of freshwater fish which are monitored in selected locations in the rivers in the Czech Republic.
