Vztah polymorfizmů genu pro eozinofilní peroxidázu k patogenezi alergických nemocí

Association of single nucleotide polymorphisms in the eosinophil peroxidase gene with etiopathogenesis of allergic diseases

Abstract:

ANOTACE Práce je zaměřena na analýzu genu pro eosinofilní peroxidázu (EPO), kationický protein uvolňovaný ze specifických granul aktivovaných eosinofilů. Eosinofilní granulocyty hrají centrální úlohu při obraně organizmu proti parazitům, účastní se ale také efektorové složky alergického zánětu. Variabilita genu pro EPO byla zkoumána pouze ojediněle, převážně u dědičné deficience eosinofilní peroxidázy, která je velmi vzácným onemocněním. Vztahem polymorfismů tohoto genu ke klinickému fenotypu alergických chorob se dosud zabývaly pouze dvě japonské práce, které prokázaly asociaci mezi určitými variantami tohoto genu a alergickou rýmou u nemocných přecitlivělých na pyl cedru. V jiných populacích nebyl tento gen prozatím studován ani z hlediska výskytu různých polymorfismů, ani ve vztahu k alergiím. Tato pilotní studie je prvním pokusem o vyhledání polymorfismů v genu pro EPO typických pro českou populaci a o rozbor jejich vztahu k různým projevům alergických nemocí a jejich intermediárním fenotypům. Analýza genu pro EPO byla provedena moderními metodami molekulární biologie (polymerázová řetězová reakce (PCR), heteroduplexní analýza (HA-PCR), analýza konformačního polymorfismu jednořetězců (SSCP-PCR), sekvencování) formou case-control studie. Celkem bylo nalezeno pět polymorfismů, 1710T/C, 1710T/CTCC, 2648T/C, 3096A/G, 3979A/G (Arg220Arg), v genu pro eosinofilní peroxidázu. Vybrané polymorfismy byly poté studovány analýzou polymorfismu délky restrikčních fragmentů (PCR-RFLP) ve vztahu k alergickému fenotypu. Našli jsme významnou asociaci jak polymorfismu 3979A/G (Arg220Arg) v exonu 6, tak i variant 2648T/C v intronu 3 a 3096A/G v intronu 5 s manifestací alergického onemocnění, případně s hodnotami celkových IgE a počty eosinofilů v krvi. Naše výsledky svědčí o tom, že varianty genu pro eosinofilní peroxidázu pravděpodobně hrají významnou roli v etiopatogenezi alergických onemocnění v naší populaci.

Abstract:

ANNOTATION This work is based on the study of analysing the eosinophil peroxidase (EPO) gene, which is a cationic protein released from specific granules of activated eosinophils. Eosinophil granulocytes play a central role in protection of organisms against parasites. They are regarded also as key effector cells in allergic inflammation. Variabily of the gene for the EPO has been previously examined only in a few isolated cases and mainly in hereditary eosinophil peroxidase deficiency which is a very rare disease. The association of polymorphisms in the eosinophil peroxidase gene to a clinical phenotype of allergic diseases was a centre of only two studies performed in Japan which managed to show the association between explicit variations of the particular gene and the japanese cedar pollinosis. This gene has not been further tested or traced either in other population samples or from the point of view of occurance of different polymorphism and their associations to allergies. This pilot study is thus the first known attempt to locate the polymorphisms in the EPO gene typical for the Czech population and to analyse their associations to different allergic reactions of diseases and their intermediary phenotypes. The analysis of the EPO gene was performed by the latest methods of molecular bilology such as polymerase chain reaction (PCR), heteroduplex analysis (PCR-HA), single-strand conformation polymorphism analysis (PCR-SSCP), and DNA sequencing in the form of case-control study. Polymorfisms with higher frequency were consequently tested with the help of PCR-restriction fragment length polymorphism analysis (PCR-RFLP) in its association to allergic fenotype. In total, 5 polymorphisms, 1710T/C, 1710T/CTCC, 2648T/C, 3096A/G, 3979A/G (Arg220Arg), in the eosinophil peroxidase gene were found. The chosen polymorphisms were conseuently studied in the form of PCR-restriction fragment length polymorphism analysis (PCR-RFLP) in association to the allergic phenotype. There has also been found a significant association of both the polymorphisms 3979A/G (Arg220Arg) in exon 6 and the variation 2648T/C in intron 3 and 3096A/G in intron 5 with the manifestation of the allergic disease, also with the values of the total IgE and amounts of levels of eosinophils in blood. These results give the evidence for the fact that the gene variations for the eosinophil peroxidase most probably play a significant role in etiopathogenesis of the allergic diseases in our population.
