Use of avoidance tests forevaluation of solid samples toxicity

Abstract:

Kontaktní biotesty jiţ nyní představují významný nástroj hodnocení reálné toxicity půdních matric a do budoucna jejich význam pravděpodobně ještě vzroste. Nicméně akutní a subletální testy jsou často nepraktické díky jejich délce trvání, náročnosti i ceně. Jako vhodná alternativa nebo důleţitý doplněk se jeví testy únikového chování s půdními bezobratlými. Základním principem těchto testů je schopnost půdních organismů rozeznat stresující prostředí a snaha z něho uniknout. Cílem mé práce bylo: (a) zvládnutí a optimalizace reprodukčního testu s Eisenia fetida (b) zvládnutí a optimalizace únikového testu s E. fetida (c) naměření efektů vytěţených 30 sedimentů z projektu NAZV oběma postupy (d) vyhodnocení výsledků a porovnání výpovědní hodnoty obou testů. Výsledky ukázaly velkou korelaci mezi počtem juvenilů a počtem kokonů u subletálního testu. V únikovém testu byla velká variabilita dat a nebyla objasněna příčina únikovosti, ale ani preferencí určitých vzorků. Poměr ředění kontaminovaných sedimentů čistou půdou, který je stanoven na 1:3 (kontaminovaná : čistá), se na základě výsledků této práce, jeví jako dostatečný. Vzorky, které vychází z obou testů jako nejméně vhodné pro organismy se u kaţdého testu různí. Ačkoliv není moţné srovnání citlivosti jednotlivých testů, testy únikového chování vyšly jako praktičtější.

Abstract:

Contact bioassays are currently very important tool for evaluation real toxicity of solid materials, moreover, their importance will probably increase. However, acute and reproduction tests are often not practical due to their duration, material intensity and cost. Avoidance tests with soil invertebrates could be an alternative or important addition to the sublethal tests because of their short duration, low cost and sensitive relevant endpoint – behavior. Basic principle of these tests is ability of soil invertebrates to recognize stress environment and attempt to avoid it. The aims of my study were: (a) managing and optimizing sublethal reproduction test with earthworms Eisenia fetida (Oligochaeta) (b) managing and optimizing avoidance dual chamber bioassays with E.fetida (c) measuring effects of dredged sediments mixed with uncontaminated soil in rate 1:3 (sediment : control soil) to earthworm E.fetida by both tests (d) analyzing results and comparing them. Results show high correlation between sublethal endpoints, quantity of juvenils and quantity of cocoons. Data from avoidance behavior tests were relatively variable. There were many replicates where organisms even preferred test soil to control soil. The real reason of such behavior in these tests was not clarified. Ratio for dilution of sediments with control soil appears to be sufficient. Samples which have the worst indication are different for both used tests. Although the results are not completely clear, the avoidance behavior test can be confirmed as an useful additional tool for assessment ecotoxicity for terrestrial organisms.
