SYNTÉSA THIOLOVÝCH SLOUČENIN U MODELOVÝCH ROSTLIN EXPONOVANÝCH TOXICKÝM TĚŽKÝM KOVŮM

EXPRESSION OF THIOL COMPOUNDS AT MODEL PLANTS TREATED WITH TOXIC HEAVY METALS

Abstract:

Obranyschopnost rostlin je klíčová pro jejich existenci v prostředí, kde je ohrožují biotické a abiotické stresové faktory. Během vývoje se rostliny vybavily velkým množstvím obranných drah, při kterých vznikají a zpětně se regenerují sloučeniny, které umožňují rostlinám aktivně ovlivňovat jak stresové faktory, tak i následky vzniklé jejich působením. Olovo dokáže velice nepříznivě ovlivnit celkový stav a metabolismus exponovaných rostlin. Rostliny se tudíž brání syntézou obranných látek – thiolových sloučenin. Nejvýznamnější jsou glutathion a fytochelatiny. Díky nim jsou schopny eliminovat toxický kov a také se bránit proti procesům, které jsou toxickým kovem indukované. V experimentu byly rostliny slunečnice roční exponovány toxickému komplexu Pb EDTA (0, 100, 250, 500 µM) po dobu sedmi dnů. Nejprve bylo nezbytné zvolit a optimalizovat vhodnou analytickou metodu pro stanovení thiolových látek. Byla zvolena a optimalizovaná technika vysoko účinné kapalinové chromatografie s elektrochemickou detekcí, která se ukázala jako dostatečně senzitivní (detekční limity v řádech jednotek až stovek pg . ml-1) a selektivní, a proto byla použita pro analýzu rostlinných vzorků. Ze získaných výsledků je patrné, že všechny stanovené thioly (cystein Cys, redukovaný glutathion GSH, oxidovaný glutathion GSSG a fytochelatin PC2) byly aktivně zapojeny do obranného mechanismu exponovaných rostlin slunečnice roční. Množství GSH se do třetího dne expozice velmi výrazně snížilo u všech koncentrací chelátu a poté byl pozorován do pátého dne pozvolný nárůst na hladinu kontroly, což bylo pravděpodobně způsobeno celkovým špatným stavem rostlin a spotřebou GSH na syntézu PC2. Navíc bylo sledováno ovlivnění jak růstu, tak i množství chlorofylu u exponovaných rostlin. V tomto ohledu olovo ukázalo vysokou fytotoxicitu už od počátku expozice a výrazně snížilo množství chlorofylu a tím ovlivnilo fotosyntézu. Tím došlo pravděpodobně k omezení přísunu energie pro redukci GSSG, jehož množství proto do konce experimentu narůstalo. Bylo také zajímavé sledovat množství volného olova akumulovaného do podzemní a nadzemní části exponovaných rostlin. Během sedmi dnů expozice dokázaly rostliny slunečnice do své nadzemní části akumulovat nejméně 37,4 mg . kg-1 sušiny olova. Hyperakumulátor olova je definován jako rostlina obsahující 1 mg olova na kg sušiny. Tento předpoklad rostlina splnila. Na základě všech získaných poznatků je tedy možno tvrdit, že zkoumaná rostlina je schopna se ucházet o své místo mezi hyperakumulátory olova v životním prostředí.

Abstract:

The protective function of plants is crucial for their existence at the enviroment, where they are threatening by biotic and abiotic stress factors. During their evolvement they have created the large number of the protective pathes, which allow the plants to influence both the stress factors as well as the consequences resulted from its activity. The lead can negatively influence the entire condition and metabolism of exposed plants. Plants are defending themselves by synthesis of protective substances – thiol compounds. The most important are glutathion and phytochelatins. Thanks to them they are able to eliminate toxic metal as well as to defend themselves against toxic metal induced processes. In this experiment were the plants of sunflower exposed to the toxic chelat of Pb EDTA (0, 100, 250, 500 µM) for seven days. First of all it was necessary to choose and optimize suitable analytical method for detection of thiol compounds. The chosen and optimized technics was high performance liquid chromatography with elecrochemical detection, which has showen to be highly sensitive (the detection limit in 1 – 100 pg . ml-1), and selective, and therfore has been used for plant samples analysis. The obtained results have shown, that all detected thiols (Cys, reduced glutathion GSH, oxidized glutathion GSSG and phytochelatin PC2) were actively involved into protective mechanisms of exposed plants. The amount of GSH has been extremely decreasing to third day in every concentrations of chelat and after this there have been observed gradual increasing to the level of control to the fifth day, what was caused by deteriorated condition of plants and entire consumption of GSH for synthesis of PC2. Additionally there was observed influencing of the growth, as well as the amount of chlorophylle in exposed plants. In this perspective the lead has shown high phytotoxicyty from the beginning of the experiment and significant amount of chlorophylle and thus have been the photosynthesis influenced. And thus have presumably occured the reducing of energy intake for GSSG reduction, which amount therfore to the end of the experiment has increased. It was also interesting to monitor the level of free lead accumulated in roots, stem and leaves of exposed plants. During the seven days of exposition were the plants able to accumulate at least 37,4 mg . kg-1. The lead hyperaccumulator is defined as a plant which contains 1 mg . kg-1 of lead in solids. The plant has fulfilled this expectation. At the basis of the obtained results is possible to claim, that the tested plant is able to be ranked between the hyperaccumulators of lead in the environment.
