Antiapoptotic function of mutant p53

Abstract:

Mutace v genu TP53 nevede pouze ke ztrátě schopností proteinu fungovat jako nádorový supresor, ale mutantní protein získává řadu vlastností napomáhající tvorbu nádorů. Tedy mutantní proteiny nejsou inertní, ale ovlivňují významně transkriptom a fenotyp nádorových buněk. Kromě podpory proliferace, diferenciačního bloku, či angiogeneze a ivazibility mezi onkogenní vlastnosti patří také léková rezistence a inhibice apoptózy. Právě posledně zmíněné vlastnosti jsou spojeny s mechanizmem zahrnujícím vazbu proteinů na DNA. A to buď přímou strukturně selektivní vazbu mutantní p53 k regulačním oblastem na či vazbu zprostředkovanou transkripčními faktorůy (např. E2F1, VDR, SP1 atd.). Významná je v této souvislosti také úloha interakce mutantních p53 s dalšími členy proteinové rodiny p63 a p73 na proapoptotické vlastnosti p63 a p73. V rámci bakalářské práce budou shrnuty poznatky z této oblasti s důrazem na roli strukturně specifické interakce mutantní p53 s DNA.

Abstract:

Mutations in the TP53 gene does not lead only to the loss of the ability of protein function as a tumor suppressor, but mutant protein receives a number of properties conducive to tumor formation. Thus, the mutant proteins are not inert, but significantly affect the transcriptome and phenotype of tumor cells. In addition to promoting proliferation, differentiation block or between angiogenesis and oncogenic properties also include drug resistance and inhibition of apoptosis. It is the latter features are associated with the mechanism involving the binding of proteins to DNA. And either direct structurally selective binding mutant p53 in the regulatory regions or binding-mediated transcription factors (eg, E2F1, VDR, SP1, etc.). Significant in this context also the role of the interaction of mutant p53 protein with other family members p63 and p73 in the proapoptotic properties of p63 and p73. The thesis will be summarized findings from this area with emphasis on the role of structure-specific mutant p53 interaction with DNA.
