The Effect of Temperature and Sodium Chloride Content on the Growth of Arcobacter Butzleri

Abstract:

Teoretická část diplomové práce charakterizuje rod Arcobacter, zabývá se jeho výskytem, vlastnostmi, jeho patogenitou, metodami inhibice a především fyzikálními a chemickými faktory využívané pro růst a přežívání rodu Arcobacter spp. Práce byla zaměřená na růstové charakteristiky A. butzleri v laboratorním médiu při teplotách 25, 30 a 37 °C a s různým obsahem chloridu sodného. Bylo pracováno s celkovými koncentracemi 1, 2, 3 a 4% NaCl v CASO bujónu. Dále byla práce zaměřená na schopnosti buněk adaptace na vybrané koncentrace NaCl. Na závěr byl zkoumán vliv vybraných cukrů a AMK v prostředí 2 a 3% NaCl na růst A. butzleri. V této práci bylo zjištěno, že A. butzleri nejlépe rostl při 25 a 30 °C. Za přídavku solí A. butzleri rostl nejlépe v bujónu s koncentrací 0,5 ? 2% NaCl. V přítomnosti 3 a 4% NaCl byly naměřeny velmi nízké ?nulové? absorbance (hodnoty od 0 do 0,060), ale po vyočkování na CASO agary byl zjištěn růst arkobakterů i v těchto koncentracích po 24 hod. Dále bylo zjištěno, že po růstu A. butzleri v živném médiu s koncentrací 3% NaCl, došlo k adaptaci buněk na prostředí s vyšším osmotickým tlakem. Vyšší hodnoty absorbancí s delší dobou růstu byly zaznamenány v živném médiu s obsahem 1, 2, 3 a 4% NaCl. Zároveň u koncentrace 4% NaCl byl patrný zákal a naměřeny vyšší absorbance, a to především u teploty 30 °C. Na závěr byl zkoumán vliv cukrů maltóza, glukóza a aminokyselin L-histidin a L-asparagin v koncentraci 2 a 3% NaCl při 30 °C a nebyl shledán významný vliv na růst a přežívaní A. butzleri. Přidané látky nezpůsobily žádné změny v naměřených hodnotách absorbancí v živném médiu s vyšší koncentrací chloridu sodného. Tyto látky pravděpodobně nemají pro buňky A. butzleri osmoprotektivní účinek.

Abstract:

The theoretic part of this thesis characterizes the stock Arcobacter, deals with his occurrence, features, his pathogen, methods of inhibition and above all with physical and chemical factors exploited for growth and survival of the stock Arcobacter spp. The thesis was intented on vegetative characteristics A. butzleri in a laboratory medium at temperatures 25, 30 and 37 °C and with a different content of sodium chloride. Total concentrations 1, 2, 3 and 4% NaCl in CASO bouillon were used. Next, the thesis was focused on cell´s possibilities of adaptation on chosen concentrations NaCl. Lastly, the influance of chosen sugars and AMK in environment 2 and 3% NaCl on growth A butzleri was reviewed. In this thesis was found out that the A. butzleri grew best at 25 and 30 °C. Thanks to the addition of salts A. butzleri grew best in the bouillon with concentration 0,5 ? 2% NaCl. In the presence of 3 and 4% NaCl very low, ? zero" absorbances (values from 0 to 0,060) were measured, but after vaccination on CASO agars the growth of arcobacters was also found out in these concentrations after one-day cultivation. Further was ascertained, that after the growth of A. butzleri in nutritive medium with the concentration 3% NaCl, cell´s adaptation on environment with higher osmotic pressure was realized. Higher values of absorbances with longer time for growth were recorded in the nutritive medium with content 1, 2, 3 and 4% NaCl. At the same time in the concentration 4% NaCl a cataract was perceptibled and also higher absorbances were measured, namely at temperature 30 °C. Lastly, the influence of sugars maltose, glucose and amino - acids L- histidine and L- asparagine in concentration 2 and 3% NaCl at 30 °C was reviewed and a significant influence on growth and survival of A. butzleri was not found. Added substances did not cause any changes in measured values of absorbances in nutritive medium with higher concentration of sodium chloride. These substances probably do not have osmoprotective effect on cells A. butzleri.
