Verification of possible determination of xanthophyll cycle pigments from the extract for the enzyme Rubisco assay

Abstract:

Tato bakalářská práce je zaměřena na ověření nové metodiky, která umožňuje sta-novení pigmentů xantofylového cyklu z extraktu pro stanovení aktivity enzymu Rubisco. Jako teoretické předpoklady k experimentální části jsou uvedeny struktura, funkce a vlast-nosti fotosyntetických pigmentů, zvláště karotenoidů, a možnost jejich degradace během analýz. Dále je stručně popsán průběh a význam xantofylového cyklu, vlastnosti a funkce enzymu Rubisco a spektrofotometrická metoda stanovení jeho aktivity. V posledním oddílu teoretické části jsou shrnuty vlastnosti chemikálií užitých při analýze enzymu Rubisco a fy-zikálně chemické vlastnosti rozpouštědel. Metodiky stanovení aktivity enzymu Rubisco a stanovení obsahů fotosyntetických pigmentů v rostlinném pletivu byly propojeny a použitelnost této modifikace byla testována na rostlinách ječmene jarního (Hordeum vulgare L. cv. Bonus). Pro hodnoty deepoxidač-ních poměrů (DEPS) a poměrů pigmentů xantofylového cyklu nebyly zjištěny signifikantní rozdíly mezi metodikou využívající extraktu enzymu Rubisco a standardní metodou extrak-ce pigmentů z čerstvého rostlinného materiálu. Nezbytnost použití korekčních faktorů byla statisticky prokázána v případě absolutních hodnot obsahů pigmentů neoxantin, violaxantin, anteraxantin 1, lutein, chlorofyl b a chlorofyl a a v případě poměrů obsahů pigmentů chloro-fylu a a chlorofylu b. Nově zavedená metodika byla aplikována v experimentech sledující krátkodobý vliv zvýšené koncentrace oxidu uhličitého na fotosyntetický aparát buku lesního (Fagus sylvati-ca) a smrku ztepilého (Picea abies) v průběhu dne. Byla zjištěna odlišná odezva DEPS těchto rostlinných druhů. Smrk ztepilý pod vlivem zvýšené koncentrace CO2 vykazuje v poledních hodinách vyšší hodnoty DEPS oproti rostlinám vystaveným přirozené koncent-raci CO2, zatímco u buku lesního je tomu naopak.

Abstract:

This bachelor thesis is aimed at verify the new method which make possibility to de-termine content of the xanthophylls pigment from the extract for Rubisco activity determi-nation. There is introduced the structure, function and characteristics of photosynthetic pigments (carotenoids especially) and a possibility of their degradation during analysis as a theoretical assumption for the experimental part. The importance of the xanthophylls cycle, characteristics and function of Rubisco and the spectrophotometrical determination of the Rubisco activity are presented too. The characteristics of chemical compound for Rubisco enzyme analysis and physico-chemical properties of many solvents are summarize. The methods for the Rubisco activity and determination of the photosynthetic pig-ments were interconnect. The applicability of this modification was tested using plants of spring barley (Hordeum vulgare L. cv. Bonus). There wasn´t found any significant differ-ences for de-epoxidation state of xanthophylls (DEPS) between both methods (the method used the Rubisco enzyme extract and the standard method of pigment extract from the fresh plant material). In the case of the absolute value of the pigment content (neoxanthin, an-theraxanthin, lutein, chlorophyll b, chlorophyll a and chlorophyll a/b ratio) the necessity of employment of correction factors was significantly demonstrated. The newly established method was applied in the experiments focused on the short-therm effect of higher CO2 concentration to the photosynthetic apparatus of European beech (Fagus sylvatica) and Norway spruce (Picea abies) during the day. There was de-tected different reaction of DEPS between this plant species. Norway spruce under the higher CO2 concentration shows in the midday hours the higher DEPS compared to the plants in natural CO2 concentration. In the case of European beech the results was opposite.
