Analysis of SF3B1 gene and protein in chronic lymphocytic leukemia patients

Abstract:

Chronická lymfocytární leukémie (CLL) je nejčastějším typem leukémie v západním světě. Během posledních několika let byly objeveny rekurentně mutované geny, jejichž inaktivace v patogenezi CLL nebyla předtím předpokládána. Největším překvapením byla identifikace mutací v genu SF3B1 (sestřihový faktor 3 podjednotka 1), který je důležitou komponentou sestřihového aparátu. Tato diplomová práce se zabývá analýzou genu a proteinu SF3B1 u pacientů s CLL. Identifikace mutací v genu SF3B1 byla prováděna pomocí Sangerova sekvenování a analýza proteinové hladiny byla stanovována westernovým přenosem. Mutační screening genu SF3B1 byl následně vyhodnocen ve vztahu k proteinové hladině SF3B1 a k molekulárně cytogenetickým datům pacientů. Metodika byla aplikována na souboru 80 pacientů s diagnostikovanou CLL, u nichž bylo nalezeno celkem 19 patologických sekvenčních změn (mutací). Výsledky mutačních analýz byly statisticky zhodnoceny v kontextu s výsledky molekulárně cytogenetických vyšetření a byly nalezeny statisticky významné souvislosti mezi výskytem mutací v genu SF3B1 a mutacemi v genu ATM a rovněž inaktivací dráhy ATM/p53. Zjištěné poznatky podtrhují význam defektů v genu SF3B1 pro patogenezi CLL a také jejich významnost pro prognózu pacientů.

Abstract:

Chronic lymphocytic leukemia (CLL) is the most common type of leukemia in the Western world. Recurrently mutated genes, whose inactivation in CLL pathogenesis had not been previously anticipated, have been discovered during the last few years. The most surprising was the identification of mutations in the gene SF3B1 (splicing factor 3 subunit 1), which is an important component of the spliceosome. This diploma thesis deals with the analysis of SF3B1 gene and protein in patients with CLL. Identification of mutations in the gene was performed using Sanger sequencing and analysis of the protein level was done using western blot. The outputs of the mutation screening were subsequently correlated with the SF3B1 protein level and molecular cytogenetic data of patients. The methodology was applied to a cohort of 80 patients diagnosed with CLL, in which 19 pathological sequence variations (mutations) were detected. Results of the mutational analysis were statistically evaluated in the context of the molecular cytogenetic data. Statistically significant associations were observed between the presence of mutations in SF3B1 and (a) ATM mutations, and (b) ATM/p53 pathway inactivation. The findings underscore the importance of SF3B1 mutations for CLL pathogenesis and also for patients´ prognosis.
