Genotoxicity potentials of air pollutants in the high mountain habitats

Abstract:

Niektoré vzdušné polutanty sú schopné transportu na dlhé vzdialenosti od svojho zdroja. Vysoké pohoria západných Karpát predstavujú bariéru pre tieto látky. Táto doktorská štúdia bola zameraná na stanovenie genotoxického potenciálu celkovej zmesi vzdušných polutantov a biomonitoring kontaminácie životného prostredia v horských a alpínskych biotopoch. Hodnotené boli chemické látky schopné diaľkového transportu. Hlavnou skupinou analyzovaných polutantov boli ťažké kovy (najmä Pb, Cd). Okrem toho bola štúdia zameraná na overenie a optimalizáciu použitých metód pre tieto ekosystémy, overenie vhodnosti použitých druhov bioindikátorov (ich akumulačnej schopnosti) a vyhodnotenie faktorov ovplyvňujúcich mieru genotoxického účinku a akumulácia sledovaných chemických látok v rastlinných tkanivách. Na zistenie miery genotoxicity prostredia bola použitá biologická metóda založená na mikroskopickej analýze peľu - Test abortivity peľových zŕn. Chemické analýzy rastlinných vzoriek boli uskutočnené pomocou AAS a XRF spektrometrie. Calluna vulgaris, Pinus mugo, Pinus sylvestris, Polytrichum commune, Pleurozium schreberi boli použité ako bioindikátory.

Abstract:

Some air pollutants are capable of long-range transport from their source. The high mountains of the Western Carpathians represent a barrier for these substances. This doctoral study was focused on the determination of the genotoxic potential of the total mixture of air pollutants and biomonitoring of environmental contamination in the mountain and alpine habitats. Chemical substances capable of long-range transport were evaluated. The main group of analyzed pollutants were heavy metals (mainly Pb, Cd). In addition, the study aimed to verify and optimize the used methods for these ecosystems, to verify the suitability of bioindicator species (their accumulation capacity) and to evaluate the factors affecting the level of genotoxic effect and accumulation of monitored chemicals in plant tissues. A biological method based on the microscopic analysis of pollen grains was used to determine the degree of genotoxicity of the environment - Pollen grain abortion assay. Chemical analyses of plant samples were performed using AAS and XRF spectrometry. Calluna vulgaris, Pinus mugo, Pinus sylvestris, Polytrichum commune, Pleurozium schreberi were used as bioindicators.
