Detection of tetraspanins in the male reproductive tract and spermatozoa

Abstract:

Jedním z klíčových dějů během oplození je fúze spermie a vajíčka. Aby u savců mohlo dojít k momentu splynutí gamet, jsou nezbytné interakce proteinů, které zahrnují také proteiny z tetraspaninové nadrodiny. Ačkoli je v posledních letech vliv tetraspaninů na mechanismy reprodukce a jejich zapojení v nich předmětem intenzivního zkoumání, prozatím není zcela přesně znám. Nicméně víme, že pro mechanismy oplození je klíčové zapojení tetraspaninových členů a jejich schopnost asociovat s jinými povrchovými proteiny a vytvářet tím tetraspaninovou síť v gametách.Tetraspaniny CD9, CD81 a CD151 byly a stále jsou intenzivně studovány na plazmatické membráně vajíčka mnoha savčích druhů. Jejich výzkum probíhá v posledních letech taktéž na spermiích, nicméně pozornost byla zatím věnována jen několika savčím druhům. Proto bylo cílem naší práce tyto tetraspaniny detekovat na membránách spermií a v reprodukčním traktu (1) prasat, u nichž byly studovány jen zřídka, a (2) hřebců, u kterých prozatím studovány vůbec nebyly.Rešeršní část této práce je zaměřena na shrnutí dosavadních informací o CD9, CD81 a CD151. Předmětem experimentální části práce byla jejich detekce a lokalizace v reprodukčním traktu a spermiích.Pomocí nepřímé imunofluorescenční mikroskopie jsme tyto tetraspaniny lokalizovali v reprodukčních tkáních a v epididymálních a ejakulovaných spermiích. Protein CD9 byl pomocí specifických protilátek lokalizován ve spermatických buňkách semenotvorného kanálku a Leydigových buňkách varlat kanců. U hřebců byla lokalizace CD9 prokázána pouze ve spermatických buňkách. V kančím i hřebčím nadvarleti jsme CD9 lokalizovali ve spermiích uvnitř kanálku nadvarlete, ve tkáni a také v sekrečním epitelu. Zatímco výsledky detekce CD9 u epididymálních hřebčích spermií byly negativní, u kanců jsme CD9 lokalizovali na vnitřní akrozomální membráně a u ejakulovaných kančích spermií navíc ještě v oblasti krčku. V oblasti akrozomu a krčku jsme detekovali CD9 u hřebčích ejakulovaných spermií. Získané výsledky jsme ověřili metodou Western blot a následné imunodetekce proteinů v extraktech epididymálních a ejakulovaných kančích a hřebčích spermií, kde byly detekovány proteiny o přibližných molekulových hmotnostech 26 a 28 kDa.V apikální oblasti akrozomu ejakulovaných kančích spermií jsme pomocí nepřímé imunofluorescenční mikroskopie lokalizovali tetraspanin CD81, který jsme rovněž imunodetekcí zaznamenali v extraktech ejakulovaných kančích spermií. U hřebců byl z extraktů epididymálních spermií detekován CD81 o molekulové hmotnosti 28 kDa. Zbývající výsledky detekce a lokalizace CD81 u kančího a hřebčího modelu byly negativní.A konečně byl také lokalizován tetraspanin CD151 ve spermatických buňkách semenotvorného kanálku kančího varlete. Pozitivní reakce protilátky byla rovněž viditelná v sekrečním epitelu kanálku kančího nadvarlete, kde byly zřetelné jednotlivé sekreční váčky. V intersticiální tkáni hřebčího varlete a ve tkáni a sekrečním epitelu kanálku hřebčího nadvarlete jsme zaznamenali pouze slabou reakci protilátky proti CD151. Výsledky imunolokalizace u hřebčích epididymálních spermií byly negativní. Naproti tomu jsme pozorovali pozitivní vazbu protilátky na protein CD151 u epididymálních kančích spermií, kde jsme CD151 lokalizovali v subekvatoriálním segmentu. U ejakulovaných kančích spermií byl CD151 detekován v oblasti akrozomu a taktéž v subekvatoriálním segmentu. V ekvatoriálním segmentu byl CD151 lokalizován u hřebčích ejakulovaných spermií. Imunodetekci jsme provedli rovněž u tetraspaninu CD151. V extraktech epididymálních kančích spermií jsme detekovali protein o molekulové hmotnosti 50 kDa a u hřebců protein

o hmotnosti 49 kDa. Detekce CD151 z extraktů kančích ejakulovaných spermií byla negativní. Na druhou stranu u hřebců byl detekován protein o molekulové hmotnosti 38 kDa. Detekce CD151 ve vyšších molekulových hmotnostech ukazuje na glykosylované formy tohoto proteinu.Z našich výsledků je patrné, že lokalizace CD9, CD81 a CD151 v samčím reprodukčním traktu a ve spermiích prasat a koní je podobná lokalizaci, jež byla zjištěna u spermií jiných savčích druhů (např. u býka). Můžeme z toho usuzovat, že role těchto tří tetraspaninů na spermiích prasat a koní je velmi podobná funkci u jiných savčích druhů. Významné je především jejich zapojení v interakcích a fúzi spermie s vajíčkem. Do těchto interakcí se zapojují právě díky své schopnosti asociovat s jinými proteiny, a vytvářet tak organizovanou tetraspaninovou síť, pomocí které jsou schopny ovlivňovat mechanismy během oplození.

Abstract:

One of the key events during fertilization is the fusion of sperm and egg. In mammals, interactions of tetraspanin superfamily proteins as well as other proteins are necessary for gamete fusion. Although the influence of tetraspanins on the mechanisms of reproduction and on their involvement in those mechanisms has been the subject of intensive research in recent years, it is not fully known yet. However, we know that the involvement of tetraspanin family members and their ability to associate with other surface proteins, and thus form a tetraspanin network in gametes, is crucial for fertilization mechanisms.The tetraspanins CD9, CD81, and CD151 have been intensively studied on the plasma membrane of eggs of many mammalian species. Their research has also been conducted on sperm in recent years, but so far only few mammalian species have received attention. Therefore, this thesis aimed to detect these tetraspanins on sperm membranes and in the reproductive tract of (1) pigs, in which they have been rarely studied, and (2) stallions, in which they have not been studied at all.In the research part of this thesis, we focused on summarizing the existing information about CD9, CD81, and CD151. The subject of the experimental part of the thesis was their detection and localization in sperm and reproductive tract.Using indirect immunofluorescence microscopy, we localized these tetraspanins in reproductive tissues and in epididymal and ejaculated sperm. Using specific antibodies, the CD9 protein was localized in spermatic cells inside the lumen of seminiferous tubules and Leydig cells of boar testes. In stallions, the localization of CD9 was demonstrated only in sperm cells. In boar and stallion epididymis, we localized CD9 in sperm inside the epididymal duct, in the tissue, and in the secretory epithelium. The results of CD9 detection in epididymal stallion sperm were negative; in boars, we localized CD9 on the inner acrosomal membrane, and in ejaculated boar sperm, we localized it on the inner acrosomal membrane and in the neck area. In the acrosome and neck area, we detected CD9 in stallion ejaculated sperm. We verified the obtained results by Western blot and immunodetection of proteins from extracts of epididymal and ejaculated boar and stallion sperm, where proteins with approximate molecular weights of 26 and 28 kDa were detected.In the apical region of the acrosome of ejaculated boar sperm, we localized tetraspanin CD81 using indirect immunofluorescence microscopy. We also recorded CD81 by immunodetection from ejaculated boar sperm extracts. In stallions, CD81 with a molecular weight of 28 kDa was detected from epididymal sperm extracts. The remaining results of CD81 detection and localization in the boar and stallion models were negative.Moreover, the tetraspanin CD151 was localized in spermatic cells inside the lumen of seminiferous tubules of boar testes. A positive antibody reaction was also visible in the secretory epithelium of the boar epididymal duct, where individual secretory vesicles were distinct. We observed only a weak anti-CD151 antibody response in the interstitial tissue of the stallion testes and in the tissue and secretory epithelium of the stallion epididymis. In stallion epididymal sperm, there were negative immunolocalization results. In contrast, we observed positive antibody binding to CD151 protein in epididymal boar sperm, where we localized CD151 in the subequatorial segment. In ejaculated boar sperm, CD151 was detected in the acrosome and also in the subequatorial segment. In the equatorial segment, CD151 was localized in ejaculated stallion sperm. We performed immunodetection on tetraspanin CD151, too. From extracts of epididymal boar sperm, we detected a protein with a molecular weight of 50 kDa; and, in stallions, a protein with a molecular weight of 49 kDa. of 38 kDa was detected. Detection of CD151 at higher molecular weights indicates glycosylated forms of this protein.Our results show that the localization of CD9, CD81, and CD151 in the male reproductive tract and in the sperm of pigs and horses is similar to that found in the sperm of other mammalian species (e.g., bull). Based on that, we may assume that the role of these three tetraspanins in pig and horse sperm is very similar to their function in other mammalian species. Their involvement in interactions of sperm and egg and in the sperm-egg fusion is especially important. They engage in these interactions precisely because of their ability to associate with other proteins, and thus create an organized tetraspanin network, through which they are able to influence mechanisms during fertilization.
