Use of chaperones in the preparation of medically relevant recombinant proteins

Abstract:

Bakalářská práce se zabývá využitím chaperonů v přípravě medicinálně významných rekombinantních proteinů. Po stručném úvodu, který se věnuje vzniku proteinů, následuje kapitola zabývající se chaperony obecně. Tato kapitola vysvětluje, k čemu v buňce slouží a představuje základní komplexy chaperonů nacházející se v prokaryotické buňce. Další část práce je věnována rekombinantním proteinům, konkrétně historii a expresním systémům, ve kterých je možné rekombinantní proteiny produkovat. Následně jsou rozebrány jednotlivé části expresního vektoru, tedy například promotor, selekční marker, afinitní značka a další komponenty. V této kapitole je také uvedeno využití chaperonů k získání proteinů z inkluzních tělísek, do kterých se proteiny často formují při expresi v cizím hostiteli. Na konci práce jsou uvedeny kmeny bakterie Escherichie coli, které se často používají k výrobě rekombinantních proteinů. V samotném závěru jsou poté uvedeny jednotlivé proteiny, které se mohou vyrábět rekombinantní technologií v prokaryotické buňce.

Abstract:

The bachalor thesis is focused on usage of chaperones in the preparation of medically significant recombinant proteins. After the brief introduction about protein synthesis, the chapter about chaperones in general comes. This chapter explains the role of chaperones in the cell and introduces basic complexes of chaperones located in the prokaryotic cell. Next part of this thesis is devoted to recombinant proteins especifically to the history and expression systems, in which recombinant proteins can be produced. Then there is a part about each component of expression vectors, such as promoter, selection marker, affinity tag and other components. In this chapter is also mentioned usage of chaperons to obtain proteins from inclusion bodies into which proteins often form after expression in foreign host. At the end of this thesis strains of bacteria Escherichia coli, which are commonly used for production of recombinant proteins are described. At the very end individual proteins, which can be produced by recombinant technology in prokaryotic cells are described.
