Structure and function of phage endolysine phi812 and their recombinant forms

Abstract:

Rostoucí výskyt bakteriálních patogenních kmenů je poslední dobou záležitostí celosvětového významu a z rychlého nárůstu rezistence bakterií vůči antibiotikům se stal mimořádně palčivý problém. Zdá se, že možnosti tvořit stále nová a účinnější antibiotika jsou pomalu vyčerpány a výskyt složitě léčitelných infekcí se dramaticky zvyšuje po celém světě. Bakterie se pomalu stávají rezistentní vůči většině nebo všem dostupným antibiotikům. A to je důvod, proč se vědci a farmaceutické společnosti zaměřují na alternativní způsoby léčby. Jedním z možných náhrad antibiotik jsou virové enzymy endolysiny, které jsou schopny rozštěpit buněčné stěny bakterií. V této práci se věnujeme studiu endolysinu bakteriofága phi812. Tento fág je schopný infikovat široké spektrum stafylokoků, MRSA (methicillin-rezistentní S. aureus) kmeny nevyjímaje. Naším cílem je připravit funkční enzym a poskytnout jeho detailní biochemickou charakterizaci včetně návrhu pravděpodobného reakčního mechanismu.

Abstract:

An increasing amount of bacterial pathogenic strains are now a reality of global significance, and the rapid emergence of antibiotic resistance to bacterial pathogens has become a particularly persistent problem. It seems that the possibilities for new and more efficient antibiotics being created are slowly exhausted, and the incidence of difficult-to-treat bacterial infections has dramatically increased worldwide. Bacteria slowly become resistant to most or all available antibiotics. So that is the reason why the researchers and pharmaceutical companies have focused on alternative ways. One of the possible replacements for antibiotics are viral enzymes, namely endolysins, which are able to disrupt the cell wall of bacteria. In this thesis we study the endolysin of bacteriophage phi812. The phage phi812 infects a wide range of staphylococci, the MRSA (methicillin-resistant S. aureus) included. Our goal is to prepare a functional enzyme and to provide its detailed biochemical characterization, including a proposal of a possible reaction mechanism.
