Immunocytochemistry utilizing labels based on photon-upconversion nanonoparticles

Abstract:

Rakovina prsu zůstává pro ženy závažným zdravotním problémem, který vyžaduje přesnou detekci biomarkerů pro včasné odhalení a lepší výsledky léčby. Imunohistochemie (IHC) a imunocytochemie (ICC) jsou běžně používané bioanalytické metody pro detekci antigenů ve vzorcích tkání nebo buněčných kultur v diagnostice i výzkumu. Tyto metody jsou založeny na protilátkách spojených s detekčními značkami, což umožňuje vysoce specifickou vizualizaci antigenů. Typicky používané značky jsou obvykle enzymy nebo fluorescenční barviva, často však nejsou dostatečně citlivé. Jako potenciální alternativa pro řešení tohoto problému se nabízejí foton-upkonverzní nanočástice (UCNPs). Tyto nanokrystaly dopované lanthanoidy dokáží absorbovat dva nebo více nízkoenergetických fotonů a následně emitovat foton s vyšší energií. Díky nízkoenergetické excitaci je minimalizována autofluorescence, což snižuje interference optického pozadí a vede k citlivější detekci. Cílem této práce bylo vyvinout vysokokontrastní metodu ICC s využitím značek na bázi UCNPs pro detekci klinicky významných biomarkerů, jako je HER-2 (lidský epidermální růstový receptor 2) u karcinomu prsu. Za tímto účelem byly otestovány čtyři konjugáty UCNPs se streptavidinem (SA-UCNPs). Konjugáty byly nejprve charakterizovány metodami dynamického rozptylu světla (DLS) a analýzy trajektorie nanočástic (NTA). Následovalo modelové stanovení využívající hovězí sérový albumin konjugovaný s biotinem (BSA-bt), kde byla ověřena jejich funkčnost. Poté byly konjugáty testovány ve stanovení ULISA (upconversion-linked immunosorbent assay) pro detekci lidského sérového albuminu (HSA) a srovnány se stanovením ELISA (enzyme-linked immunosorbent assay). Při tomto srovnání dosahovala ULISA lepší výsledky, s přibližně 4× nižším limitem detekce a přibližně 6× vyšším poměrem signálu ku pozadí (S/B). Nakonec byly konjugáty UCNPs použity pro ICC značení HER-2, kde pomocí SA-UCNPs dopovaných Tm3+ bylo dosaženo specifické detekce, což dokazuje jejich potenciál pro citlivou detekci klinických biomarkerů. Nakonec byla otestována digitální detekce jednotlivých UCNPs pomocí epiluminiscenčního mikroskopu; tato metoda významně zvýšila poměr S/B a prokázala tak účinný přístup pro zvýšení kontrastu při upkonverzní detekci.

Abstract:

Breast cancer remains a serious health problem for women worldwide, demanding precise biomarker detection for early identification and improved therapy outcomes. Immunohistochemistry (IHC) and immunocytochemistry (ICC) are widely utilized bioanalytical methods for detecting antigens in tissue samples or cell cultures, used mainly in diagnostics or research. These methods are based on antibodies coupled with detection labels, enabling highly specific antigen visualization. However, the typically used labels for these methods, such as enzymes or fluorescent dyes, are often not sensitive enough. To address this issue, photon-upconversion nanoparticles (UCNPs) offer a potential alternative. These lanthanide-doped nanocrystals can absorb two or more low-energy photons and subsequently emit a higher-energy photon. Thanks to low-energy excitation, autofluorescence is minimized, reducing optical background interference and leading to more sensitive detection. This thesis aimed to develop a high-contrast ICC method utilizing UCNP-based labels for detecting clinically relevant biomarkers, such as HER-2 (human epidermal growth receptor 2) in breast cancer. For this goal, four streptavidin-conjugated UCNPs (SA-UCNPs) were tested out. Conjugates were firstly characterized by dynamic light scattering (DLS) and nanoparticle tracking analysis (NTA). This was followed by a model assay using biotin-conjugated bovine serum albumin (BSA-bt) to verify their functionality. The conjugates were then tested in an upconversion-linked immunosorbent assay (ULISA) for the detection of human serum albumin (HSA) and compared with an enzyme-linked immunosorbent assay (ELISA). In this comparison, ULISA proved to be superior, reaching approximately 4× lower limit of detection and approximately 6× higher signal-to-background ratio (S/B). Finally, SA-UCNPs were used for ICC labeling of HER-2, where specific detection was achieved using Tm3+-doped conjugate, demonstrating their potential for sensitive detection of clinical biomarkers. Furthermore, a digital readout of individual UCNPs was tested out using an epiluminescence microscope, which significantly increased S/B ratios, demonstrating efficient approach to enhance contrast in upconversion detection.
