Isolation and characterization of gram-positive-derived extracellular vesicles and monitoring of their intestinal-cell uptake <I>in vitro</I>

Abstract:

Extracelulární vezikuly (EV), membránově derivované váčky obalené lipidovou dvojvrstvou, jsou uvolňovány většinou známých typů buněk, včetně eukaryotických a bakteriálních. V posledních letech se výzkum zaměřuje na jejich roli v mezibuněčné komunikaci, díky jejich schopnosti přenášet řadu bioaktivních molekul do cílových buněk na krátké i dlouhé vzdálenosti. Tato diplomová práce se zaměřuje na EV produkované gram-pozitivním bakteriálním kmenem Lacticaseibacillus rhamnosus CCM7091 (Lb-EV), který byl původně izolován z lidského gastrointestinálního traktu a patří k druhu potenciálně prospěšných bakterií. V lidském gastrointestinálním traktu probíhá velmi intenzivní komunikace mezi hostitelem a střevní mikrobiotou a předpokládá se, že EV mohou hrát významnou roli. Cílem práce bylo izolovat, charakterizovat a fluorescenčně zaznačit Lb-EV, aby bylo možné sledovat jejich interakce a s využitím vybraných inhibitorů objasnit mechanismy jejich vstřebávání do cílových buněk. Výsledky této diplomové práce ukázaly, že Lb-EV vykazují heterogenní složení a používají více cest internalizace do epiteliálních buněk s největším příspěvkem kaveolin dependentní endocytózy. Byla také prokázána přítomnost LTA, potenciálního markeru EV z gram-pozitivních bakterií, který může přispívat k tomu, že jsou Lb-EV rozpoznávány TLR2/TLR6 receptory. Tento výzkum je klíčový pro rozšíření našich znalostí o EV pocházejících z gram-pozitivních bakterií, které jsou v porovnání s gram-negativními bakteriemi méně prozkoumané. Detailní charakterizace těchto Lb-EV a analýza mechanismů jejich absorpce mohou významně přispět k porozumění mikrobiální komunikace v gastrointestinálním traktu člověka a mohou nabídnout nové možnosti pro cílené terapeutické využití.

Abstract:

Extracellular vesicles (EV), membrane-derived vesicles encapsulated by a lipid bilayer, are released by most known cell types, including both eukaryotic and bacterial cells. Recent research has focused on their role in intercellular communication, owing to their ability to transport a variety of bioactive molecules to target cells over both short and long distances. This diploma thesis focuses on EV produced by the Gram-positive bacterial strain Lacticaseibacillus rhamnosus CCM7091 (Lb-EV), originally isolated from the human gastrointestinal tract and classified as a potentially beneficial bacterium. Intense communication occurs between the host and the gut microbiota within the human gastrointestinal tract, and EV are hypothesized to play a significant role in this process. This study aimed to isolate, characterize, and fluorescently label Lb-EV to monitor their interactions and, by using selected inhibitors, to elucidate the mechanisms of their uptake by target cells. The results of this thesis demonstrated that Lb-EV exhibits a heterogeneous composition and utilizes multiple pathways for internalization into epithelial cells, with caveolin-dependent endocytosis being the most prominent. The presence of lipoteichoic acid (LTA), a potential marker of Gram-positive bacterial EV, was also confirmed, which may contribute to the recognition of Lb-EV by TLR2/TLR6 receptors. This research is pivotal in advancing our understanding of EV derived from Gram-positive bacteria, which are comparatively less explored than those from Gram-negative bacteria. The comprehensive characterization of these Lb-EV and the examination of their absorption mechanisms significantly contribute to our understanding of microbial communication in the human gastrointestinal tract and may open pathways for novel therapeutic applications.
