Expression and acumulation of dehydrins in Opium poppy

Abstract:

Tato práce shrnuje dosavadní informace o dehydrinech a jejich akumulaci při různém vystavení stresu. K expresi a akumulaci dehydrinů dochází nejčastěji pod vlivem abiotických stresů, které jsou z důvodu extrémních klimatických změn čím dál rozšířenější. Abychom předešli ztrátám výnosů a úhynu rostlin, potřebujeme vypěstovat rostliny, které odolají těmto nepříznivým podmínkám. Modelovou rostlinou pro moji práci je mák setý, u kterého jsem popsala možné mechanismy suchovzdornosti z výsledné sekvenace RNA dvou jeho kontrastních odrůd. Vybrala jsem nejodlišněji exprimované geny a vyobrazila je pomocí heatmapy v programu R. Dalším cílem mé práce bylo pomoci s optimalizací metodiky stanovení dehydrinů. Izolované dehydriny jsem stanovila pomocí metody vertikální gelové elektroforézy, elektroforetického přenosu biomakromolekul na membránu a imunochemické detekce proteinů. Výsledky mé práce mohou dále sloužit jako podklad pro podrobnější studie dehydrinů.

Abstract:

This work summarizes existing information about dehydrins and their accumulation in various stress exposure. Expression and accumulation of dehydrins occurs most often under the influence of abiotic stresses, which are increasingly widespread due to extreme climate change. To avoid loss of yield and death of plants, we need to grow plants which can withstand these unfavorable conditions. The model plant for my work is the Opium poppy, with which I have described possible mechanisms of dry-resistance from the resulting sequencing RNA of two of its contrast varieties. I chose the most differentially expressed genes and demonstrated them using the heatmap in R. Another point of my work was to help with optimalization of dehydrin determination methodology. I determined isolated dehydrins using the method of vertical gel electrophoresis, electrophoretic transfer of biomacromolecules to the membrane and immunochemical detection of proteins. The results of my work may be useful as a basis for more detailed studies of dehydrins.
