Development of Analytical Methods for Study of Azaarenes and Azaarones in Environmental Matrices

Abstract:

Tato disertační práce je zaměřena na vývoj nových analytických metod pro stanovení heterocyklických látek, azaarenů (PANHs) a jejich degradačních produktů azaaronů (OH-PANHs). Tyto látky nejsou v současnosti ani systematicky ani intenzivně monitorovány v životním prostředí v porovnání s polycyklickými aromatickými uhlovodíky (PAHs). Toxicita azaarenů v životním prostředí je v důsledku jejich nízkých koncentrací v prostředí a toxických efektů uváděna jako relativně nízká. Toxicita azaaronů v prostřední zatím nebyla systematicky studována, ale v posledním desetiletí bylo publikováno několik studií uvádějících koncentraci azaaronu vyšší v porovnání s azaareny a toxicitu azaaronů srovnatelnou s PAHs. Tato disertační práce si klade za cíl popsat vývoj a optimalizaci metod pro měřeni azaarenů a jejich degradačních produktů azaaronů v říčních a mořských vzorcích. Tyto metody by měly doplnit soubor metod pro hodnocení celkové kontaminace prostředí polyaromatickými látkami. Vyvinutá metoda umožňující souběžnou analýzu 2 a 3 jádrových azaarenů a azaaronů ve vzorcích půdy a sedimetů je kombinací extrakce pomocí horkého rozpouštědla a hmotnostní spektrometrie s fotoionizací. Extrakce na tuhou fázi (SPE) byla úspěšně testována pro použití při analýze vodných vzorků. Mikroextrakce (SPME) byla testována také, ale jejímu širšímu použití brání zejména malý extrakční povrch a nízká hodnota distribučního koeficientu, která v tomto případě způsobuje nízkou citlivost metody. Robustnost, citlivost a přesnost nových metod byla prokázána. Využití nově vyvinutých metod bylo prokázáno při terénních experimentech. Metody byly úspěšně testovány a aplikovány v monitorovací studii pokrývající oblast Wadenského moře v Nizozemí. Výsledky potvrdily fakt, že koncentrace azaronů v prostředí jsou srovnatelné, nebo vyšší v porovnání s koncentrací azaarenů. Poměr mezi koncentrací azaarenů a azaaronů v prostředí byl výrazně ovlivněn industriálními aktivitami v regionu a intenzitou degradacetěchto látek v oblasti.

Abstract:

This dissertation thesis aims to develop new analytical methods for measurement of aromatic heterocyclic compounds azaarenes (PANHs) and their degradation products azaarones (OH-PANHs). Nowadays PANHs and OH-PANHs are not systematically and intensively monitored in the environment compared to polycyclic aromatic hydrocarbons (PAHs). Toxicity of azaarenes in the environment is expected to be relatively low because of their low concentration and toxic effects. Toxicity and occurrence of azaarones in the environment have not been systematically studied yet but in last decades several studies reporting concentrations of azaarones higher than concentration of azaarenes and their toxicities in fact comparable with the often monitored PAHs have been published. In the present work, I intend to describe the development and optimization methods of measurement of azaarenes and their degradation products, azaarones, in environmental samples collected from rivers and the sea. The method should complement the other analytical methods of evaluation of the total environmental contamination by polycyclic aromatic compounds. The newly developed method for soil and sediment matrices combines warm Soxhlet extraction with tandem mass spectrometry using atmospheric pressure photo ionization for simultaneous measurement of 2 and 3 ring azaarenes and azaarones structures. Solid phase extraction (SPE) has been successfully tested in this study as an extraction method for water matrices. Solid Phase Microextraction (SPME) has also been tested as an extraction method but a small extraction surface and calculated low distribution coefficients for PANHs limit their use in sampling and in this case also caused low sensitivity of the method. Robustness, sensitivity and accuracy of the new methods have been proven. Potential impact of the method has been shown in a field study. The method has been successfully tested and applied in a monitoring study in the Waden Sea in the northern part of the Netherlands. The results confirm that the concentrations of azaarones are comparable or even higher than azaarenes. The ratio between concentrations of azaarenes and azaarones highly depended on industrial activities in the region and on degradation activities in the area.
