Expression of genes influencing boar taint and its relation to castration

Abstract:

Cílem disertační práce bylo prokázat vliv kastrace na hladinu mRNA exprese genů CYP2E1, CYP1A2, CYP2A19, HSD3B, HSDB17B7, SULT1A1, SULT2A1, KRT8 a ATP5B a jejich vztah se složkami kančího pachu androstenonem, skatolem, indolem a protilátkami specifickými pro vakcínu Improvac u českého komerčního hybrida prasat (velké bílé x landrase (samice) x duroc (samec)). Prasata byla rozdělena na skupiny nekastrovaných (NC), chirurgicky kastrovaných (SC), imunologicky kastrovaných poražených 8 (IM8) nebo 15 (IM15) týdnů po druhé dávce vakcíny Improvac a na skupinu samic (GI). Exprese mRNA v játrech, měřena kvantitativní real-time polymerázovou řetězovou reakcí, se signifikantně lišila mezi kontrolní skupinou (NC) a ostatními skupinami u genů CYP2E1, CYP1A2 a CYP2A19. Hladina mRNA genu HSD3B se signifikantně lišila mezi kontrolní skupinou a skupinami IM8, IM15 a GI. Genová exprese SULT1A1 se lišila mezi kontrolní skupinou a skupinami SC, IM8 a GI. V případě genu SULT2A1 byl signifikantní rozdíl jen mezi kontrolní skupinou a skupinou IM8. U genů KRT8 a ATP5B nebyl pozorován žádný rozdíl v expresi. Negativní korelace mezi androstenonem a hladinou mRNA u genů CYP2E1, CYP2A19 a SULT1A1 naznačují inhibici genové exprese. Signifikantní rozdíl mezi skupinami IM8 a IM15 byl pozorován pro protilátky specifické pro Improvac, což naznačuje pokles protilátek v čase. Také jsme pomocí polymerázové řetězové reakce a Sangerova sekvenování analyzovali jednonukleotidové polymorfismy u genů CYP2E1 (g.12007C>A, c.1422C>T, c.1423A>G), CYB5A (g.165901487del G, g.165901767 T>C, g.165902078 C>T, AFO16388:c.-G>T) a HSD3B (g.165262 G>A) a jejich korelaci s androstenonem, skatolem a indolem. Žádný z těchto polymorfismů nebyl signifikantně korelován s látkami kančího pachu v námi zkoumané populaci prasat.

Abstract:

The aim of this study was to prove the influence of castration on mRNA expression level of genes CYP2E1, CYP1A2, CYP2A19, HSD3B, HSDB17B7, SULT1A1, SULT2A1, KRT8 and ATP5B and its relationship with boar taint compounds androstenone, skatole and indole and Improvac-specific antibodies in Czech commercial hybrid (Large White x Landrase (sow) x Duroc (boar)). The pigs were separated in groups of entire male (NC), surgically castrated (SC), immunocastrated by Improvac slaughtered 8 (IM8) and 15 (IM15) weeks after the second dose and gilts (GI). Gene expression of CYP2E1, CYP1A2 a CYP2A19 genes in liver measured by real-time chain polymer reaction was significantly different between control group (NC) and all other groups. For HSD3B the expression level was significantly different between control group and IM8, IM15 and GI. Gene expression of SULT1A1 was significantly different between control group and SC, IM8 and GI. In the case of SULT2A1 the gene expression differed significantly only between IM8 and control group. We have not observed any significant difference in gene expression in genes KRT8 and ATP5B. Negative correlations between androstenone and mRNA levels of CYP2E1, CYP2A19 and SULT1A1 suggest that gene expression is suppressed. Significant differences for Improvac-specific antibodies between IM8 and IM15 groups were observed, indicating decrease of antibodies over time. We have also analysed single nucleotide polymorphisms of genes CYP2E1 (g.12007C>A, c.1422C>T, c.1423A>G), CYB5A (g.165901487del G, g.165901767 T>C, g.165902078 C>T, AFO16388:c.-G>T) a HSD3B (g.165262 G>A) by polymerase chain reaction and sanger sequencing and their correlation with androstenone, skatole and indole, but we have not observed any significant correlations between polymorphisms and boar taint compounds.
