Molecular Profile of Selected Drug Biotransformation Enzymes, Transporters and Transcription Factors in Non-Obese Rat Models of Metabolic Syndrome

Abstract:

Tato disertační práce se zabývá studiem změn v expresi a aktivitě vybraných enzymů biotransformace, transportérů a transkripčních faktorů v játrech a tenkém střevě u ne-obézních potkaních modelů metabolického syndromu. Metabolický syndrom představuje komplexní patologický stav charakterizovaný poruchami metabolismu lipidů a glukózy, hypertenzí a chronickým nízkostupňovým zánětem, což jsou faktory schopné ovlivňovat farmakokinetiku léčiv a metabolismus endogenních látek. Cílem práce bylo komplexně zhodnotit, jak jednotlivé komponenty metabolického syndromu (hypertriglyceridémie, hypertenze, chronická exprese lidského C-reaktivního proteinu a hormonální deficit na-vozený ovariektomií) ovlivňují expresi a funkci klíčových enzymů 1. a 2. fáze biotrans-formace léčiv, transkripčních faktorů a vybraných ABC transportérů. Experimentální část práce byla provedena na potkanech kmenů Wistar, HHTg, SHR, SHR-CRP a na ovariektomizovaných samicích Wistar. V rámci experimentální části byla analyzována relativní exprese mRNA vybraných forem CYP, enzymů 2. fáze biotransformace (UGT, SULT, GST a COMT), ABC transportérů a jaderných receptorů v jaterní a intestinální tkáni. Současně byla stanovena relativní enzymová aktivita vybra-ných forem CYP v jaterních mikrozomech. Genová exprese byla vyhodnocena metodou RT-qPCR, enzymová aktivita pomocí luminiscenčních testů a LC/MS metod, přičemž získaná data byla statisticky zpracována standardními postupy. Výsledky práce ukázaly, že jednotlivé modely metabolického syndromu vykazují odlišné a modelově specifické změny v expresi a aktivitě biotransformačních enzymů. U potkanů kmene HHTg byly v játrech zaznamenány významné změny v expresi vybra-ných forem CYP, zejména výrazné snížení exprese Cyp1a2 a signifikantní změny v ex-presi genů Cyp2d1 a Cyp2c6. U hypertenzního modelu SHR byla detekována upregula-ce genů zapojených do metabolismu kyseliny arachidonové, zatímco transgenní exprese lidského CRP vedla k divergentní regulaci konjugačních enzymů mezi játry a střevní tkání. Ovariektomie u samic Wistar způsobila pokles jaterní detoxikační kapacity a zvý-šení intestinální exprese efluxního transportéru P-glykoproteinu. Poslední část práce po-tvrdila, že suplementace silymarinem u potkanů HHTg snižuje jaterní kumulaci kyseliny fenofibrové, pravděpodobně cestou aktivace enzymů druhé fáze biotransformace. Vý-sledky přispívají k objasnění mechanismů, kterými metabolický syndrom modifikuje biotransformační systémy a farmakokinetiku léčiv.

Abstract:

This dissertation thesis focuses on the study of changes in the expression and activity of selected drug biotransformation enzymes, transporters, and transcription factors in the liver and small intestine of non-obese rat models of metabolic syndrome. Metabolic syn-drome represents a complex pathological condition characterized by lipid and glucose metabolism disorders, hypertension, and chronic low-grade inflammation, which are factors capable of influencing drug pharmacokinetics and the metabolism of endogenous substances. The aim of this work was to comprehensively evaluate how individual com-ponents of metabolic syndrome (hypertriglyceridemia, hypertension, chronic expression of human C-reactive protein, and ovariectomy-induced hormonal deficit) affect the ex-pression and function of key phase I and phase II biotransformation enzymes, transcrip-tion factors, and selected ABC transporters. The experimental part of the study was conducted on Wistar, HHTg, SHR, and SHR-CRP rat strains, and on ovariectomized Wistar females. Within the experimental part, the relative mRNA expression of selected CYP forms, phase II biotransformation enzymes (UGT, SULT, GST, and COMT), ABC transporters, and nuclear receptors was analyzed in the hepatic and intestinal tissues. Concurrently, the relative enzyme activity of selected CYP forms in hepatic microsomes was determined. Gene expression was evaluated using the RT-qPCR method, and enzyme activity was assessed via lumine-scent assays and LC/MS methods, with the acquired data being statistically processed using standard procedures. The results of the study demonstrated that individual models of metabolic syn-drome exhibit distinct and model-specific changes in the expression and activity of bio-transformation enzymes. In HHTg rats, significant changes in the expression of selected CYP forms were observed in the liver, particularly a marked decrease in Cyp1a2 expres-sion and significant changes in the expression of Cyp2d1 and Cyp2c6 genes. In the SHR model, the upregulation of genes involved in arachidonic acid metabolism was detected, whereas the transgenic expression of human CRP led to a divergent regulation of con-jugation enzymes between the liver and intestinal tissue. Ovariectomy in Wistar females caused a decline in hepatic detoxification capacity and an increase in the intestinal ex-pression of the efflux transporter P-glycoprotein. The final part of the study confirmed that silymarin supplementation in HHTg rats reduces the hepatic accumulation of feno-fibric acid, likely via the activation of phase II biotransformation enzymes. The results contribute to elucidating the mechanisms by which metabolic syndrome modifies bio-transformation systems and drug pharmacokinetics.
